Inonotus Obliquus Ethanol Extract Effects On TNF-a And IL-2 Of MFC Gastric Cancer Tumor-bearing Mice And Its Pharmacodynamics Study

Objective:To preliminarily study the pharmacodynamics of Inonotus Obliquus ethanol extracts,as well as its inhibitory effect on tumors,impact on immune organs and effect on TNF-α and IL-2 of MFC gastric cancer tumor-bearing mice.Methods:MFC gastric cancer model was established with BALB/c mice of which armpit was implanted 0.2ml of gastric cancer cell strain with the concentration of 2×107.The inhibitory effect of Inonotus Obliquus ethanol extract on MFC gastric cancer tumor-bearing mice was observed and the tumor-inhibition rate was calculated.The spleen mass,thymus gland mass,spleen index and thymus gland index were determined.The impact of Inonotus Obliquus ethanol extract on TNF-α and IL-2 in serum of MFC gastric cancer tumor-bearing mice was tested by ELISA method.The impact of Inonotus Obliquus ethanol extract on tumor cells morphology of MFC gastric cancer tumor-bearing mice was observed by HE stain.The anti-inflammatory and analgesic effects of Inonotus Obliquus ethanol extract on Kunming mice were observed by paw swelling method and acetic acid writhing method.The impact of Inonotus obliquus ethanol extract on physical strength of Kunming mice was observed in anti-fatigue experiment and anti-hypoxia experiment.Results:The inhibitory rate of Inonotus Obliquus ethanol extract from high-to-low doses on MFC gastric cancer tumor-bearing mice were respectively 53.2%,41.2%and 33.6%.The inhibitory effect was positively correlated with the concentration of Inonotus Obliquus ethanol extract.Inonotus Obliquus ethanol extract increased the spleen index and thymus gland index as well as the content of TNF-αand IL-2 of MFC gastric cancer tumor-bearing mice at different levels.Thus,the immunity of MFC gastric cancer tumor-bearing mice was improved.Inonotus Obliquus ethanol extract significantly decreased the paw swelling rate of Kunming mice.Additionally,it significantly reduced writhing times of Kunming mice in the acetic acid writhing experiment.The pain inhibition rates of the high-dose,medial-dose and low-dose group given Inonotus Obliquus ethanol extract as well as the group given 5-fluorouracil were respectively 62.78%,23.89%,5.00%and 48.89%,compared with that of blank group.Inonotus Obliquus ethanol extract increased the anti-hypoxia time,and at different levels,increased the burdened swimming time of Kunming mice,which indicates that it can enhance the physical strength of Kunming mice.This effect is positively correlated with the concentration of Inonotus Obliquus ethanol extract.Conclusion:Inonotus Obliquus ethanol extract can effectively inhibit the growth of MFC gastric cancer tumor cells and improve the immunity and physical strength of mice,thus achieving the anti-tumor effect.
