In this paper,the water extract of Inonotus obliquus was obtained by means of hot water extraction,and its components were determined.The immunological functional activity of the water extract of Inonotus obliquus was studied by animal experiments.At the same time,the preliminary toxicological evaluation of the the water extract of Inonotus obliquus was carried out by acute oral toxicity test,which provided a safe basis for the application of the water extract of Inonotus obliquus in the development of food and medicine health care products.The following conclusions can be drawn:(1)The extraction rate of polysaccharides and flavonoids in Inonotus obliquus by hot water was studied by one-factor experiment and response surface design.The optimal conditions for total extraction of flavonoids and polysaccharides from Inonotus obliquus were obtained:liquid ratio 25(v/w),extraction time 2.5 h,extraction temperature 95 ?,the conditions were verified,the actual average polysaccharide yield obtained was 1.16%,the yield of flavonoids was 3.98%,and the actual value Basically consistent with the theoretical value.(2)Through the analysis of the components in the water extract of Inonotus obliquus,the results showed that the content of polysaccharide of Inonotus obliquus extract was 4.71 mg/g,the flavone content was 14.00 mg/g,the fat was 0.43 mg/g,the crude protein content was 2.10 mg/g,the potassium content was 118.28 g/kg,the magnesium content was 166.57 mg/kg,the sodium content was 134.48 mg/kg,the manganese content was 91.03 mg/kg and the zinc content was 42.96 mg/kg,iron content of 31.18 mg/kg,copper content of 4.86 mg/kg and selenium content of 0.25 mg/kg.Meanwhile,the contents of heavy metals and microbial indicators in the water extract of Inonotus obliquus were all tested in accordance with the national food safety standards Request.(3)Mice immunosuppressed by establishing cyclophosphamide model mice were orally gavaged with different doses of the water extract of Inonotus obliquus,evaluation of the mice weight,spleen index,thymus index,the killing activity of NK cells and peritoneal macrophage phagocytosis,spleen lymphocyte proliferation capacity,glutathione peroxidase (GSH-PX),superoxide dismutase(SOD),malondialdehyde(MDA),total antioxidant capacity(T-AOC),catalase(CAT),alkaline phosphatase(AKP)content change index,immunomodulatory effect of the water extract of Inonotus obliquus was studied.The results showed that compared with the model group,the doses of 500 mg/kg,1000 mg/kg,2000 mg/kg the water extract of Inonotus obliquus could significantly increase the body weight and organ index of immunocompromised mice,Phagocytosis and proliferation of splenic lymphocytes have a good recovery,NK cell cytotoxic activity was significantly improved;on the liver and serum GSH-PX,SOD,CAT,AKP enzyme activity increased.(4)Acute oral toxicity test of the water extract of Inonotus obliquus was carried out by Horn's method.The results showed that the oral acute LD50 of the water extract of Inonotus obliquus was greater than 10.00 g/kg-BW.The survival mice showed normal activities.There was no significant difference between the control group and the experimental groups in body weight,viscera ratio,ALT,AST,BUN and CRE.Therefore,water extract of Inonotus obliquus was classified as innocuity.
