In this paper,single factor water extraction and ethanol extraction were studied.On the basis of dry extract of obliquus obliquus,through the response surface optimization method,the extraction times,the ratio of liquid to liquid,the extraction time were optimized respectively.The optimum conditions were ethanol concentration,solid-liquid ratio,extraction time,extraction times and temperature.The results of optimum extraction by water were 55 times the ratio of material to liquid,extraction time 77 min,extraction times of 4 times,and determine the optimum extraction conditions of ethanol concentration 55%,extraction time 98 min,extraction 4 times,extraction temperature of 55℃,the ratio of material to liquid 26 times volume of solvent.Secondly,the water extract and ethanol extract of ginseng to dry extract,on the basis of single factor analysis,response surface method,the water extraction conditions for solid-liquid ratio,extraction time,extraction times,screening and determine the optimum extraction conditions of optimum extraction time 63 min,extraction times is 3,the ratio of liquid to 61 as the volume of solvent.The ethanol extraction conditions for ginseng dry extract,ethanol concentration,extraction times,extraction time,extraction temperature,solid-liquid ratio,in the single factor experiment,response surface optimization,the optimum extraction conditions of ethanol concentration 66%,extraction time 3.2h,extraction 4 times,extraction temperature of 81℃,the ratio of material to liquid 26 times volume of solvent.To study the compatibility of two kinds of Chinese medicine,the Inonotus obliquus water extract and ethanol extract of ginseng mixture,water extracts and ethanol extracts were mixed.And then Inonotus obliquus and ginseng were mixed in accordance with 1:2,1:1,2:1.The contents of total polyphenols,total polysaccharides and total three terpenes were studied respectively.To analytice chemical components by HPLC-MS.To study the compatibility of 2 species of diabetes mellitus with the combination of obliquus and ginseng,the mice were randomly divided into eight groups,including model group,acarbose positive group,birch ginseng(2:1)and low dose group(150mg/kg,300mg/kg),Hua Shen(1:1)and low dose group(150mg/kg,300mg/kg),Hua Shen(1:2)and low dose group(150mg/kg,300 mg,/kg).After 4 weeks,glucose tolerance in mice were experimentalled,according to the determination of TC,TG,HDL-C,LDL-C,MDA,SOD index,effects on liver and kidney tissue pathology.The liver of diabetic model mice in SOD decreased significantly,MDA increased significantly.In both Shen Hua(2:1)high dose group,and Shen Hua(1:2)low dose group,liver SOD increased,MDA decreased significantly compared with model(P<0.01).In both Shen Hua(2:1)high dose group,Shen Hua(1:2)the low dose group TC,TC,LDL-C decreased,and at the same time,the level of HDL-C and the model was increased significantly(P<0.01)which improved the blood lipid level.By observing the biopsy of liver and kidney pathology,the compatibility of two traditional Chinese medicines can repair tissue damage in varying degrees. |
