Objective:To observe the effect of Chinese medicine Wugong Baidu Decoction on serum IL-17 levels in K5-STAT3C mouse,analyse the MPASI score of before and after administration and front and back difference,and take correlation analysis between serum IL-17 content and MPASI score,to explore the effect of Wugong Baidu Decoction on Th17 conduction pathway in the treatment of psoriasis.Methodes:To selecte 25 K5.STAT3C transgenic psoriasis model mice(180-220g),all were male,and divide them into 5 groups,each group 5 only.They were low dose group,middle dose group,high dose group,western medicine control group of methotrexate and normal saline blank control group,respectively given to the Wugong Baidu Decoction concentrations of 1.48g/ml,2.97g/ml,5.94g/ml,methotrexate suspension 0.11mg/ml and normal saline.After 1 h at last hole,to make injection anesthesia in each group of transgenic mice,get 4ml blood from the abdominal aorta,and extract PBMC flow cytology in 2 hours,take detection of serum IL-17 content by ELISA.Meanwhile,the PASI scoring method for evaluating the skin lesions of human psoriasis was further improved and optimized to become the MPASI score used to evaluate the area and severity of psoriasis skin lesions in mice.The correlation between serum IL-17 content and MPASI score was also analyzed.Result:1.Compared with blank control group,The serum levels of IL-17 was different in middle dose groups of Wugong Baidu Decoction(p<0.05);It was also significant difference in methotrexate western medicine control group and high dose group of Wugong Baidu Decoction(P<0.01).2.The MPASI score of methotrexate western medicine control group and high dose group of Wugong Baidu Decoction changed the largest,meaning highes score of ? MPASI;The lower were low dose group,middle dose group of Wugong Baidu Decoction;The change of normal saline blank control group was the least,meaning the lowest score of?MPASI.3.A double variate regression analysis was conducted to determine the serum IL-17 content in mice and the MPASI change in mice before and after administration(? MPASI),finally,the linear regression equation was obtained as follows:Y =25.396-1.025X,correlation coefficient y=-0.884,P=0.000<0.05,the difference was statistically significant,and linear correlation regression equation was established.Conclusion:1.The Wugong Baidu Decoction can reduce the level of serum IL-17 in mice,and show a certain quantity-effect dependence.2.The Wugong Baidu Decoction can improve the MPASI score of mice and promote the repair of psoriasis skin lesions,within a certain range,the higher the concentration of centipede,the more obvious the improvement of skin lesions in mice.3.The change of mouse skin lesion score before and after intervention of Wugong Baidu Decoction had a negative correlation with the level of IL-17 in serum of mice MPASI.4.Wugong Baidu Decoction maybe could cure psoriasis by regulating Th17 cell transduction pathway.
