Objective: To research the quality analysis of Cryptolepis buchananii Roem.et Schult as well as its chemical resistance efficacy of liver injury.Methods:(1)In accordance with the 2015 edition of the Chinese Pharmacopoeia Appendix pharmaceutical quality analysis and research methods degree of Cryptolepis buchananii Roem UV,IR and TLC identification,as well as water,ash and extract check.(2)Determination of caffeic acid in Cryptolepis buchananii Roem by high performance liquid chromatography.(3)HPLC fingerprinting of the extract of Cryptolepis buchananii Roem was studied.(4)Taking the different polarity extracts of the Cryptolepis buchananii Roem as the research object,a chemical liver injury model was established by using D-GalN to observe the changes of serum alanine aminotransferase(ALT),aspartate amino(AST)and liver homogenate superoxide dismutase(SOD),malondialdehyde(MDA)and liver histopathological changes.Results:(1)The identification methods for ultraviolet,infrared and thin layers of Cryptolepis buchananii Roem medicinal materials were determined.The stem of Cryptolepis buchananii Roem had less than 13.01% moisture,the total ash was no higher than 15.32%,the acid-insoluble ash was no higher than 0.55%,and the extract was not higher than 11.75%;Cryptolepis buchananii Roem medicinal leaves,moisture is not higher than 10.37%,total ash is not higher than 23.14%,acid insoluble ash is not higher than 3.96%,the extract is not higher than 19.92%.(2)The HPLC method was used to determine the content of caffeic acid in Cryptolepis buchananii Roem.The results of methodological study showed that all the RSD values were less than 3%.The contents of caffeic acid in 10 ginkgo herbs were 0.0965% ~0.4772%.(3)A common pattern of the fingerprints of the effective parts of the anti-hepatic injury of the extract of Cryptolepis buchananii Roem was established,and a total of 13 common peaks were calibrated.The similarity between the 10 batches of production maps and the control fingerprints was above 0.9.(4)Cryptolepis buchananii Roem can reduce the serum ALT,AST activity(P <0.05),increase the SOD activity(P <0.05)and decrease the MDA level(P <0.05).Conclusion:(1)The experiment conducted an initial study on the quality analysis of Cryptolepis buchananii Roem medicinal plants,which could provide a scientific basis for its quality control and quality standard formulation in the future.(2)The HPLC method was established for the determination of caffeic acid in Cryptolepis buchananii Roem.The method is accurate and reliable,has good linearity and high precision,and can be used for the quality evaluation of Cryptolepis buchananii Roem.(3)The common pattern of the fingerprints of the effective parts of the anti-hepatic injury of the extract of Cryptolepis buchananii Roem vine medicinal herbs was established.The method is characterized by strong characteristics,accuracy and reliability,methodological investigations are all in line with the requirements,and can provide a basis for the quality evaluation of Cryptolepis buchananii Roem.(4)The extract of Cryptolepis buchananii Roem has a protective effect on chemical liver injury induced by D-GalN in mice.
