Objective: Intracerebral hemorrhage is one of the stroke,about 15 to 20% of stroke,morbidity and mortality rate,and can be left with severe neurofunctional defect symptoms,leading to a low quality of life in the patient.After intracerebral hemorrhage,secondary brain tissue damage is the key factor affecting the prognosis and prognosis of patients,and the formation of inflammatory factors plays a decisive role in it.Mainly through secondary brain injury after intravascular red blood cells to release into the parenchymal destruction,release of inflammatory factors such as TNF-a and IL-1?,triggering the activation of inflammatory pathways,produce inflammation causes the destruction of blood-brain barrier and a large number of the death of brain cells.Brain tissue around hematoma after intracerebral hemorrhage in the expression of HO-1 raised obviously,and can be related signal pathways regulating numerous inflammatory factors,so as to exert its anti inflammatory reaction of nerve protective effect.Therefore,this research object is to people around the hematoma after intracerebral hemorrhage group,HO-1,TNF-a and IL-1? in the hematoma after intracerebral hemorrhage in the surrounding tissue inflammation not phase study and explore the dynamic expression at the same time,in order to provide new theoretical basis for the treatment of cerebral hemorrhage.Methods: Object of this study was to line of 32 cases of hypertension cerebral hemorrhage admitted in neurosurgery craniotomy hematoma removal of patients,take around hematoma organization defined as hematoma by 1 cm brain tissue as experimental group,and take a surgical approach path from hematoma in normal brain tissue as control group.According to the time of onset,the time of the brain tissue samples were divided into ?6h group,6-24 h group,24-72 h group and >72h group.By RT-PCR detection,Western blot analysis and Immunohistochemical staining,the dynamic expression of Ho-1,IL-1? and TNF-a in different time periods after intracerebral hemorrhage were detected.Results: The detection of Ho-1,IL-1? and TNF-a in Westemblot showed that the control group had weak expression,and the experimental group was less than 6h,and the expression of 6-24 h was significantly increased,and the peak of 24-72 h was reached,and a slight decrease in >72h.There was no significant difference between the experimental group and the control group(P>0.05).The time period between 6-24 h,24-72 h and >72h was significantly higher than that in the control group,and the difference was significant(P<0.05).The results of RT-PCR in the detection of Ho-1,IL-1? and TNF-a were low expression in the normal brain tissues of the control group,and the expression of Ho-1 was the highest among the three groups.The expression of the experimental group was higher than 6h,and the expression of 6-24 h was increased.The expressions of 24-72 h were significantly increased,and basically reached the peak.In >72h,the expression of Ho-1,TNF-am RNA in the brain was still increasing,and the expression of IL-1? was slightly decreased.The expression of the experimental group was significantly higher than that in the control group,and there was significant statistical difference(P<0.05).The results of Immunohistochemical staining showed that there were a small number of Ho-1,IL-1? and TNF-a positive cells in the control group,and the expression of the experimental group was less than 6h after cerebral hemorrhage,no significant difference compared with the control group(P>0.05).6-24 h expression begins to rise,24-72 h expression significantly increased,and reached a peak,>72h after a slightly downward trend,the experimental group compared with control group in 6-24 h,24-72 h,>72h were higher than control group,including within 24-72 h and >72h group when a significant difference compared with control group(P < 0.01)..Conclusion(s): HO-1 expression in brain tissue around hematoma after intracerebral hemorrhage began to increase,and reached the peak,after 2-3 days its anti inflammatory reaction of nerve protective effect,its mechanism may be inhibitory activity of the NF-k B HO-1 reduce downstream IL-1? and TNF-a expression.Brain tissue around hematoma after intracerebral hemorrhage in inflammation,TNF-a,IL-1? is involved in important factors of inflammatory reaction around the hematoma after intracerebral hemorrhage,cerebral hemorrhage within 6h begin to express,namely peaked 2-3 days,after a downward trend,and the expression of IL-1? results are basically consistent with the TNF-a expression,suggests the interaction between the two kinds of inflammatory factor expression.
