Objective To investigate the effect of long non-coding RNA7 regulated by c-Myc on the proliferation of SCC3 cells in tongue squamous cell carcinoma,and to find a novel molecular target for tongue squamous cell carcinoma.Methods Four lentiviral vectors pLKO.I-shc-Myc-ctrl,pLKO.1-shc-Myc,Flag and Flag-c-Myc were constructed and transfected into SCC3 cells respectively.Western Blot was used to detect the expression of c-Myc protein in the transfected cells;qRT-PCR was used to detect the expression of lncRNA7 in the transfected cells.Two lentiviral vectors pLKO.1-shlncRNA7-ctrl and pLKO.1-shlncRNA7 were reconstructed and qRT-PCR was used to detect the expression of lncRNA7 in the transfected SSC3 cells.Effects of lncRNA7 on the growth and proliferation of SCC3 cells were detected by MTT assay and colony formation assay respectively.The localization of lncRNA7 in SCC3 cells by fluorescence in situ hybridization.Results Western Blot showed that the expression of c-Myc protein in pLKO.1-shc-Myc group was down-regulated compared with pLKO.1-shc-Myc-ctrl group;The expression of c-Myc protein in Flag-c-Myc group was higher than that in Flag group.qRT-PCR results showed that the expression of lncRNA7 in pLKO.1-shc-Myc group was lower than that in pLKO.1-shc-Myc-ctrl group;The lncRNA7 expression in Flag-c-Myc group was higher than that in Flag group;The lncRNA7 levels in pLK0.1-shlncRNA7 group was lower than that in pLK0.1-shlncRNA7-ctrl group.The results of MTT assay and colony formation assay showed that the growth and proliferation of SCC3 cells in pLKO.1-shlncRNA7 group were weakened compared with pLKO.1-shlncRNA7-ctrl group.The results of fluorescence in situ hybridization showed that lncRNA7 was located in cytoplasm.Conclusion The down-regulation of lncRNA7 regulated by c-Myc can inhibit the growth and proliferation of SCC3 cells,thus providing a new target for gene therapy of tongue squamous cell carcinoma.
