Objective To observe the effects of transfusing 60Co ?-ray irradiated blood in patients with hematologic diseases, and the effects of 60Co?-ray irradiation on whole blood T lymphocyte subsets and cytokines.Method 198 patients with hematologic diseases who were transfused irradiated blood(100 transfused irradiated aleukocytic suspending red blood cell,98 transfused irradiated apheresis platelets)were randomly selected as the irradiation group;A total of 182 patients with hematologic diseases with non-irradiated blood(83 transfused non-irradiated aleukocytic suspending red blood cell,99 transfused apheresis platelets)were selected as the control group. All patients were tested for blood routine within 24 hours before and after transfusion. The difference of RBC,Hb and PLT before and after transfusion and the incidence of adverse transfusion in both groups were compared. At the same time,8 ml of whole blood were collected from 16 healthy subjects,of which 4ml were tested for T cell subsets(CD3+CD4+?CD3+CD8+)by flow cytometry, and cytokines(IL-2?IL-4?IL-5?IL-10?TNF?IFN-?)detected by ELISA after 60Co?-ray irradiation, the remaining 4ml were tested for T cell subsets and cytokines without the irradiation. The difference of T cell subsets and cytokines before and after irradiation was compared.Result Comparing to pre-transfusion,the RBC,Hb and PLT were significantly increased after irradiated blood transfusion(p<0.01).The differences of RBC,Hb,PLT between transfusing irradiated blood and non-irradiated blood(p>0.05).There were no transfusion associated graft versus host disease within 1 month in the irradiation group. In addition,CD8+ T cells,IL-10 decreased significantly after irradiation(p<0.05).There was no significant difference between before and after irradiation of CD4+ T cells(p>0.05).Due to the fact that the levels of cytokines suah as IL-2?IL-4?IL-5?TNF?IFN-?,etc.were lower than the measured values in most samples, statistical comparisons could not be made.Due to the fact that the levels of cytokines such as IL-2,IL-4,IL-5,TNF,IFN-y,etc.were lower than the measured values in most samples, statistical comparisons could not be made. Conclusion In patients with hematological diseases,60CO ?-ray irradiated blood transfusion has the same efficacy in improving RBC,Hb,PLT as the same amount of non-irradiated blood.;and preventing the occurrence of transfusion associated graft versus host disease(TA-GVHD)to a certain extent. What's more,60Co ?-ray irradiation can inhibit the proliferation of lymphocytes and the secretion of some cytokines.
