ObjectiveThe establishment of lower limb ischemia model,the use of autologous bone marrow mesenchymal stem cells differentiate into vascular endothelial cells,combined with galectin-3 transplantation in the lower limb ischemia model to observe the effects of angiogenesis in lower limb ischemia model Feasibility of Autologous Vascular Endothelial Cell Transplantation in Treatment of Lower Limb Ischemic Diseases.Method?1?Bone marrow markers were obtained from bone marrow punctures of 16 New Zealand white rabbits and their bone marrow markers were obtained.BMSCs were isolated and purified by density gradient centrifugation and adherent culture.The third generation of bone marrow mesenchymal stem cells with stable growth characteristics were used,VEGF combined with bFGF directed differentiation into vascular endothelial cells,flow cytometry CD31 and vWF expression rate.?2?New Zealand white rabbits left lower extremity ligation of the common iliac artery ligation method to establish New Zealand white rabbits left lower limb ischemia model?3?Sixteen New Zealand white rabbits with lower extremity ischemia models were randomly divided into experimental group?n=8?and control group?n=8?.On the 16th day after establishment of ischemia model,15 points,Experimental group injected with autologous vascular endothelial cells and galectin-3,the control group injected with saline,DSA angiography after 3weeks to observe the blood vessels of ischemic limbs.?4?New Zealand white rabbits were sacrificed at the end of angiography and ischemic limbs specimens were taken for HE staining and the capillary density and the number of capillaries/myofibers were counted.Result?1?Flow cytometry detected CD31 expression rate of 97.3±0.8%,vWF expression was 95.7%±1.1%,New Zealand white rabbit bone marrow mesenchymal stem cells successfully differentiated into vascular endothelial cells.?2?DSA angiography showed that the lower extremity vessels in the experimental group were significantly more than those in the control group.?3?The result of HE staining showed that the capillary density and the number of capillaries/myofibers in the experimental group were higher than those in the control group.Conclusion?1?Rabbit mesenchymal stem cells can be differentiated into vascular endothelial cells as tissue-engineered seed cells.?2?Autologous vascular endothelial cells combined with galectin-3 can be more mediated by ischemic lower limb angiogenesis.
