Objective:To detect the level of mi R-134 and miR-92 a in people with Anti-HCV positive.By comparing them with that of the healthycontrol group,their application value in the diagnosis of HCV infection andevaluation of disease progression was initially explored.Methods:Clinical information,laboratory data(including HCV RNA andALT)and serum samples were collected from 60 people with Anti-HCV positive and 60 healthy controls.The serum total miRNAswasextracted,and the levels of miR-134 and mi R-92 a werequantified by realtime fluorescence quantitative PCR.The differencesbetween these two groups were preliminary analyzed..Res?lts:1.The basic parameter analysis of the subjects showed that there was no statistically significant difference in the gender composition between the healthy control group and the Anti-HCV positive group(P>0.05).In the age distribution,the average age of the Anti-HCVpositive group was higher than that in the healthy control group(P<0.05).The quantitative levels of ALT and HCVRNA in the Anti-HCVpositive group were significantly higher than that in the healthy control group,and the difference was statistically significant.2.The total RNA concentration of 60 serum samples was 24.8-44.6 ng/ul.In order to achievehe same amplification efficiency,the total RNA concentration of the sample was adjusted to 20ng/ul.3.Through literature review,we know that there were two main types of internal parameters selected in serum miRNA quantitative detection,namely U6 and miR-16.Both internal reference gene expression were characterized by high stability,but it was noted that there was a change in the expression of mi R-16 in the control group and the affected group.Therefore,U6 was selected in this study,and the stability of RNU6 B expression was verified.The results showed that the Ct value of U6 snRNA between the control group and the HCV antibody positive group was no significant difference(P=0.653>0.05).Therefore,Combining relevant literature we can conclude that U6 snRNA could be used as the reference gene in this experiment.4.Anti-HCV positive serum mi R-134 andmiR-92 a the expression level ofthe health control group were significantly increased,and the difference was statistically significant(P=0.012< 0.05),may be directly or indirectly promote the HCV infection of miR-134 and miR-92 a rise in expression.5.ROC(Receiver-operatorcharacteristiccurve)analysis demonstrated the diagnosis value of mi R-134 AUC =0.9867,(95%CI:0.9724-1.001,P<0.0001)and 0.8931(95%CI:0.8365-0.9496,P<0.0001).In the critical value of 6.115 x 10-5(miR-134)and 4.231 x 10-2(miR-92a),the sensitivity and specificity of miR-134 and miR-92 a in the diagnosis of HCV is 80% ? 98.33% and 61.67% ? 98.33% respectively.MiR-134 and miR-92 a as a serological indexes of HCV infection,with potential diagnostic value.6.According to the correlation statistical analysis of SPSS20.0,there was a significant positive correlation between mir-134 and mir-92 a in the Anti-HCV positive group: r=0.823,P=0.000(<0.05).7.MiR-134 has no correlation with ALT: r = 0.203,P = 0.119;There are positive correlations between mi R-92 a and ALT: r = 0.341,P = 0.008 < 0.05;MiR-134 has no correlation with HCV RNA: r=-0.015,P=0.909;MiR-92 a has no correlation with HCV RNA: r=0.083,P=0.529.Conclusion:1.Proper control of miRNA expression is essential for maintaining a steady state of the cell?lar machinery.The median expression levels of miR-134 and miR-92 a in people with Anti-HCV positive were significantly increased,and it is spec?lated that HCV infection may change the expression of miR-134 and miR-92 a either directly or indirectly.2.According to results of ROC analysis,miR-134 and miR-92 a can be used as helpf?l diagnosis markers.3.Spearman's rank correlation analysisshowed that there was positive correlation between miR-134 and miR-92 a,also between miR-92 a and ALT.However,there was no significant correlation between miR-134 and ALT,HCV RNA,also between miR-92 a and HCV RNA.All above suggested that serum miR134 and miR-92 a could be a separate diagnosis ofHCV infection and progression monitoring indications.
