Objective: to choose the 10 day of pregnancy rats treated with mifepristone(4mg/kg)method to establish a rat model of threatened abortion,intrauterine pregnancy rats by recording the bed points,the number of live births,calculate abortion number and abortion rate of tocolysis efficacy evaluation of Cuscuta ginseng tocolysis particles,pathological characteristics and determination of serum progesterone and pregnancy tissue by observing uterine decidua in rats(P)and human chorionic gonadotropin(HCG)content,from histology,to explore the mechanism of Cuscuta ginseng granules tocolysis tocolysis endocrinology perspective.Methods: 1.groups of female,male SD rat cage by 2:1 mating,the next morning the acquisition of rat pregnancy vaginal smear,vaginal plug observed under the progesterone(6mg/kg)every other day for 1 times;D,pregnant rats of E group respectively according to 35.6g/kg,the dose of 8.9g/kg to Tu ginseng granule orally tocolysis,1 times a day.Tenth days of pregnancy in A group pregnant rats,four B-E rats were administrated with mifepristone 4mg/kg according to the establishment of threatened abortion rat model of.3.specimen collection and detection data of twelfth days of pregnancy with chloral hydrate in the pregnant rats after anesthesia anatomy and blood samples were 4ml,centrifuged upper serum by ELISA.Determination of P,HCG content were recorded;each rat's bed points,the number of live births,and calculate the number of abortion and abortion rate;the rat uterine decidua with formalin fixed,paraffin embedded and made into slices,stainedwith HE,observed the morphological changes of decidual tissue under light microscope.Results:(1)pregnant rats the number of live births,the number of abortion and abortion rate: compared with the control group,the number of live births is far less than the control group,the two comparison has statistical difference(P < 0.01),while the number of abortion and abortion rate is higher than that of the control group,the difference was significant(P < 0.01),in taking mifepristone after threatened abortion rats in the model group,model was established successfully.The progesterone group,high dose group and low dose group compared with the model group,the number of live births were higher than those in the model group,and the number of abortions,abortion rates were lower than the model group,the difference was statistically significant(P < 0.05)compared with the progesterone group,The number of live births in high dose group than progesterone group(P < 0.05 statistically significant),and the number of abortions and the abortion rate is less than the progesterone group(P < 0.05 statistically significant);live births low dose group and progesterone group number,number of abortion and abortion rate consistent comparison(P > 0.05 NS).The high dose group compared with the low dose group,low dose group,the number of live births,the number of abortions and the abortion rate is less than the low dose group,the differences were statistically significant(P < 0.05).(2)of rat uterine decidua: morphological changes of rat uterine decidua after treatment were observed under light microscope.Visible: blank control groupEndometrial glands and stromal structure intact,the proportion of normal glands,morphological rules,structural integrity of villi visible under the microscope did not see a congestion,no tendency to keratinization and glandular differentiation development;except the blank control group,model group,progesterone group and Tu Shen Antai granule to high,low dose group of rat uterus decidua showed varying degrees of degenerative change in model group was the most serious,the decidual tissue structure is completely destroyed,interstitial congestion,a small amount of glands and irregular shape,and inflammatory cells infiltration.The remaining three groups of back stroke degrees are relatively minor,generally tended to be normal,only a few spots hemorrhage and exudation(3).Comparison of pregnancy rats serum P value: rats in the model group compared with the control group,the concentration of P decreased significantly,the analysis of statistical software,P < 0.01 was considered statistically significant.Progesterone group,high dose group and low dose group compared with the model group,the serum P concentrations were significantly increased,with statistical significance(P < 0.01);high dose group and progesterone group,the serum P concentration was higher than progesterone group(P < 0.05 statistically significant);no significant difference between the P concentration of rats in low dose group and progesterone group(P > 0.05 was not statistically significant).Compared with the low dose group,high dose groupThe rats of P were significantly higher than the low dose group and the difference was statistically significant(P < 0.05).(4)comparison of pregnancy rats serum HCG value: rats in the model group compared with the control group,the concentration of HCG decreased significantly,the analysis of statistical software,P < 0.01,that was statistically significant.Progesterone group,high dose group and low dose group compared with the model group,the serum HCG concentrations were significantly higher than those in the model group,with statistical significance(P < 0.01);high dose group and progesterone group,the serum HCG concentration was higher than progesterone group(P < 0.05 statistically significant);low doseNo significant difference was found between the concentration of rat HCG group and progesterone group(P > 0.05 was not statistically significant).Compared with the low dose group,high dose group,the concentration of HCG in rats was significantly higher than the low dose group and the difference was statistically significant(P < 0.05).Conclusion: 1.using mifepristone establish threatened abortion model is feasible,and the drug model has the advantages of simple operation,strong controllability,and is worthy of popularization and application of.2.Tu progesterone large-scale particles of high low dose ginseng tocolysis,can significantly improve the model of rat embryo survival,reduce the number of abortions,reduce the abortion rate,the tiger ginseng tocolysis granules dose effect is particularly significant,confirmed that the tiger ginseng granules have good effect of tocolysis Baotai.3.Ma Shen tocolysis granule can improve the decidua of threatened abortion rats damaged the adverse pregnancy outcomes,increased serum P,HCG levels,hormone levels necessary for maintenanceof normal pregnancy,forTo provide a good environment of pregnant rats,from histology and endocrinology confirms the mechanism of tocolysis pregnancy maintenance ginseng sweet tocolysis granules.
