The Correlation Of SIRT6 With Cardiovascular Disease In Rheumatoid Arthritis

Objective:The purpose of this study is to explore the correlation between silent information regulation factor 6(Sirtuin6,SIRT6)and rheumatoid arthritis associated Cardiovascular Disease(RA-CVD),Methods:1.First,collect the clinical data of RA patients.In the study,30 RA-CVD patients and 31 active RA patients were selected and 31 cases of healthy people were as control.Then,the difference of participants' clinical data among groups was analyzed.2.The participants' serum were collected.The SIRT6 expression in serum was analyzed by ELISA.The level of SIRT6 expression in every group was analyzed.3.The correlation between the SIRT6 expression level in serum with disease activity,traditional cardiovascular risk factors,non-traditional cardiovascular risk factors,and imaging in RA-CVD patients was analyzed.1)Disease activity:DAS28(Disease Activity for 28 joint indices score),RF(Rheumatoid Factor),Anti-CCP(Anti-Cyclic Citrullinated Peptide Antibodies),ESR(Erythrocyte Sedimentation Rate).2)Traditional cardiovascular risk factors:FBG(Fasting Blood Glucose),TG(Triglycerides),HDL-C(High-Density Lipoprotein Cholesterol),TC(Total Cholesterol),BMI(Body Mass Index).3)Non-traditional cardiovascular risk factors:CRP(C-reactive protein),PLT(platelet)FRS(Framingham Risk Score),D-D polymer,BNP(Brain Natriuretic Peptide),CK(Creatine Kinase),CK-MB(Creatine Kinase isoenzyme).4)Imaging:cIMT(carotid Inteima-Media Thickness),EF(Ejection Fraction),LVd(Left Ventricular diameter Diastolic period),LVs(Left Ventricular diameter Systolic period),RV(Right Ventricular diameter),LA(Left Atrial diameter).Results:1.RA-CVD patients had higher levels of ERS,Anti-CCP,RF,HDL-C,CRP,FRS,BNP and larger diameter LA,LVs,LVd and cIMT than RA-nCVD patients.2.Compared with Control group and RA-nCVD patients,the SIRT6 expression level from serum in RA-CVD patients was significantly lower.3.1)In Disease activity,the SIRT6 expression level was negatively correlated with DAS28,and has no correlation with RF,Anti-CCP and ESR.2)In the traditional cardiovascular risk factors,the SIRT6 expression level was negatively correlated with the FBG and TG in RA-CVD patients,and was positively correlated with HLD-C.But there was no correlation with TC and BMI.3)In non-traditional cardiovascular risk factors,the SIRT6 expression level was negatively correlated with and CRP,PLT,FRS and cTnl.There was no correlation between the SIRT6 expression level and D-D polymer,BNP,CK with CK-MB.4)In the imaging,the SIRT6 expression level was only negatively correlated with cIMT.There was no correlation between the SIRT6 expression level and EF,LVd,LVs,RV and LA.Conclusion:1.The SIRT6 expression level in RA patients' serum was negatively correlated with the occurrence of RA-CVD.2.SIRT6 may affect the occurrence of RA-CVD by regulating the inflammatory response,lipid and glucose metabolism,and thrombosis in RA patients,suggesting that SIRT6 may be a potential protective factor for RA-CVD...
