Objective:Detected the expression of IL-22 in bladder urothelial carcinoma and normal bladder mucosa at the protein and gene in the same time,to investigate the relationship of IL-22 expression and clinicopathological characteristics in human bladder urothelial carcinoma tissues.Metods:1.Fity tree bladder cancer tissue samples and 20 nomal bladder mucosal samples were collectd and their paraffin-embedded sections were immunohistochemically stained to indicate the expession of IL-22,the tumor cell staining strength and the positive cell proportion were used to determine the expression intensity of IL-22 in the tissue.comparative the expression differences of IL-22 in bladder urothelial carcinoma group and normal bladder mucous group.2.The real-time fluorescent quantitative PCR(RT-qPCR)and Western blot were used to detection the bladder cancer group and the normal bladder mucous group IL-22 mRNA and IL-22 protein expression,respectively to compare the bladder urothelial carcinoma and normal bladder mucous group in IL-22 mRNA and protein expression differences,further analysis of IL-22mRNA and protein expression in bladder urothelial carcinoma patients with different age,sex,tumor number,tumor size,primary/recurrence bladder urothelial carcinoma,pathological grading,Clinical staging(NMIBC/MIBC).Results:1.1 Expression of IL-22 protein in bladder urothelial carcinoma and normal bladder mucosa.1.1.1 The results of Immunohistochemical stained results showed that IL-22 positive staining was mainly located in the cytoplasm or cytoplasm,with brown-yellow granules.In the 53 cases of bladder urothelial carcinoma,there were 34 cases of IL-22 positive expression and 19 cases of negative expression.In 20 cases of normal bladder mucosal tissue,there were 7 cases of IL-22 positive expression and 13 negative expressions.The positive expression rate of the bladder urothelial carcinoma group was 64.2%(34/53),which was significantly higher than that of the normal bladder mucosa group35.0%(7/20),the difference was statistically significant(?~2=9.296,P<0.05).1.1.2 The results of Western blot test showed that the bladder urothelial carcinoma group and normal bladder mucosa group had obvious bands in24KDa,indicating that the normal bladder mucosa group and bladder urothelial carcinoma group had IL-22 protein expression.It was also found that the expression of IL-22 in the urothelial carcinoma group of the bladder was significantly higher than that in the normal bladder mucosa group(0.649±0.107 vs 0.407±0.09),the difference was statistically significant(P<0.05).The expression level of IL-22 protein in tumor diameter>3cm was higher than that in tumor diameter?3cm(0.69±0.10 vs 0.56±0.067),and the difference was statistically significant(P<0.05).The expression of IL-22protein in MBIC was higher than that in NMIBC(0.68±0.023 vs 0.55±0.052),and the difference was statistically significant(P<0.05).There were no significant correlation between the age(?65 vs>65=0.67±0.13 vs 0.62±0.72 P>0.05),sex(male vs female=0.67±0.24 vs 0.64±0.10 P>0.05),tumor number(Single tumor vs Multiple tumors=0.59±0.39 vs 0.63±0.18P>0.05),pathological grading(Low-grade vs High-grade=0.63±0.29 vs0.66±0.34 P>0.05),primary/recurrence bladder urothelial carcinoma(0.60±0.23 vs 0.67±0.42 P>0.05)of the patients with bladder urothelial carcinoma.1.2 Detected IL-22 mRNA in bladder urothelial carcinoma tissue and normal bladder mucosal tissue.The results of RT-qPCR showed that IL-22 mRNA was significantly higher in bladder urothelial carcinoma group than in normal bladder mucosa group(4.08±0.88 vs 1.01±0.48),and the difference was statistically significant(P<0.05).IL-22mRNA in the bladder tumor diameter>3cm was higher than that in the bladder tumor diameter?3cm(1.36±0.21 vs 1.07±0.19),and the difference was statistically significant(P<0.05).IL-22mRNA in MBIC was higher than that in NMBIC(0.68±0.023 vs 0.55±0.052),and the difference was statistically significant(P<0.05).There were no significant correlation between age(?65 vs>65=1.18±0.18 vs 1.03±0.25),sex(male vs female=1.04±0.21 vs 1.09±0.13),tumor number(Single tumor vs Multiple tumors=1.19±0.19 vs 1.01±0.28 P>0.05),pathological grading(Low-grade vs High-grade=1.09±0.29 vs 1.10±0.14P>0.05),primary/recurrence bladder urothelial carcinoma(1.17±0.13 vs1.04±0.29 P>0.05)of the patients who with bladder urothelial carcinoma.Conclusions:The expression of IL-22 mRNA and protein in bladder urothelial carcinoma is related to tumor size and whether it was muscular invaded,it is not related to patients age,sex,tumor number,pathology grade and primary/recurrence.Whether Il-22mRNA and protein can be use as prognostic prognostic factors for bladder urothelial carcinoma need more study.
