Objective:MicroRNA(miRNA) is a class of endogenous,non-coding 21-25 nucleotides RNA, which is discovered in recent years. The"seed region", which is the core sequence that encompasses the first 2-7 nucleotides at the 5'portion of miRNA, is essential for the specific suppression of targe mRNA. Recent studies have showed that miRNAs could be involved in the regulation of the proliferation, differentiation and apoptosis. Therefore, miRNA has been deemed to play a crucial role in the initiation and progression of human cancers, which could regulate many oncogenes and tumor suppressor genes.Bladder cancer is one of the most common urinary cancer in the worid. Altered expression of miRNAs have been reported in bladder cancer, and may play a critical part in carcinogenesis. With more and more miRNAs have been discovered in bladder cancer, miRNAs might play an important role in the diagnosis and therapy of bladder cancer.In the present study, we detected the expression of miR-34a in bladder cancer tissues and cell lines (T24) relative to their non-tumorous control using quantity real-time PCR. Moreover, we also examined the correlation between the expression of miR-34a and clinicopathological characteristics of bladder cancer. We wanted to identify the role of miR-34a in carcinogenesis and development of bladder cancer.Methods:We detected the relative expression of miR-34a in bladder cancer tissues of 42 patients compared to their matched non-tumor adjacent tissues (NATs) by RNA isolation, Poly(A) tail and real-time RT-PCR. Therefore, data analysis was performed by the relatively quantitative 2△△C t method. We also analyzed the association between miR-34a expression and clinicopathological characteristics in bladder cancer. After transfected with the mature body of miR-34a and anti-righteousness chain into T24 cell, the biology behavior were measured by MTS assay and flow cytometry. All statistical analyses were performed with SPSS v.13.0. Statistical analysis was performed using the t-test and Mann-Whitney U test analysis,and P<0.05 was considered statistically significant.Results:We found the significant low-expression of miR-34a in bladder cancer tissues relative to their NATs. Among 42 patients with bladder cancer, 26(61.9%) cases showed a significant low-expression of miR-34a. Low expression of miR-34a was associated with increased tumor size and malignant degree. The patients with low expression of miR-34a tended to have larger tumor size and higher malignant degree. The data showed that turn into miR-34a ex-bodies of T24 cell,grow speed to show Zhao to lower,expect the cell period to distribute obvious decrease in the s,and the cell Diao Wu comparison is obviously high at the matched control.Conclusions:Explicit miR-34a was significant down-regulated in bladder cancer tissues relative to their NATs. And Low-expression of miR-34a was associated with increased tumor size and malignant degree. Furthermore, MiR-34a could inhibit growth and incude apoptosis to bladder cancer cells.
