Objective:To explore the association between IL-23 receptor gene polymorphisms and risk of upper urinary calculus in Chinese by investigation of gene polymorphisms of interleukin-23 receptor in patients group and controls group,explore.Materials and methods:(1)207 patients with confirmed upper urinary calculus were allocated to this study,who were hospitalized in the Affiliated Hospital of North Sichuan Medical College between June 2016 to June 2017,and 246 healthy controls were recruited by the physical examination during the same time,excluding interference factors such as tumor,endocrine disease and autoimmune disease.(2)Polyacrylamide gel electrophoresis and Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)were used to analyze the SNP genotypes.(3)The SPSS22.0statistical software was used to analyze the data,by comparing the distribution differences between allele frequencies and genotype frequencies in the two groups,to understand the correlation of the gene polymorphisms with susceptibility of upper urinary calculus.Results:(1)Clinical characteristics were no significant difference between cases and controls.Mean age was(51.81±13.68)years for patients and(50.30±12.5)years for healthy controls,P=0.244,there was no statistical significant.135 and 72 of the cases and 151 and 95 of the controls were men and women,(?~2=0.71,P=0.399).Body mass index(BMI)between cases and controls were(23.25±3.12 vs 23.01±2.8).The random blood glucose between the cases and the controls were(7.00±3.16 vs 6.85±2.71),P>0.05 did not have significant difference.(2)There was a significant difference in the distribution of IL-23R gene polymorphism at polymorphic site rs10889677 between patients and normal individuals,allele C frequency in cases(40.1%)was significantly higher than in controls(27.2%),(P<0.001;OR,95%CI:1.79,1.335-2.37);genotype CC frequency in cases(18.4%)was also significantly higher than in controls(8.9%),(P<0.001;OR,95%CI:2.94,1.61-5.26);There was no significant difference between cases and controls in overdominant gene model(AA+CC)vs AC(P=0.14).(3)The C allele frequency of rs11465817 was significantly lower in cases(27.8%)than in controls(37.8%),(P<0.001;OR,95%CI:0.63,0.48-0.84);genotype CC frequency in patients was(7.7%)lower than in controls(15.8%),(P<0.001;OR,95%CI:0.38,0.20-0.71);There was no significant difference between cases and controls in overdominant gene model(AA+CC)vs AC(P=0.41).Conclusion:(1)The rs10889677 and rs11465817 polymorphic site of IL-23R may be correlated with the incidence of upper urinary calculus in Chinese population.(2)The frequency of CC genotype of rs10889677 was significantly increased in the patients group(18.4%)than in the controls group(8.9%),the results revealed that CC genotype appeared to have higher susceptibility to urinary stones.Compared with the A allele,the rs10889677 C allele was significantly associated with increased risk of upper urinary calculus.(3)However,the frequency of CC genotype of rs11465817 was lower in the cases(7.7%)than in the controls(15.8%),the rs11465817 C allele was lower in cases(27.8%)compared with controls(37.8%).This suggests that it might play a role in reducing urinary calculus.
