| Objective: To investigate the correlation between the single nucleotide polymorphisms(SNPs) of-152G/A in the promoter region of IL-17 A gene,7488T/C in the exon region of IL-17 F gene and asthma children,and the association with total serum Ig E(TIg E) and serum ECP levels,to study the correlation between serum ECP and asthma.Methods: 224 asthmatic children(asthma group) and 150 healthy children(control group) were recruited.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used to detect the genotypes of IL-17 A gene-152 locus and IL-17 F gene 7488 locus. The TIg E levels were determined by enzyme-linked immunosorbent assay(ELISA).Using Beckman coulter experiment system IMMAGE 800 specific protein analyzer to scattering turbidmetric method to automatically detect serum TIg E level. Sex ratio,genotype frequency and allele frequency of asthma group and control group was compared by chi-aquare test,average age,ECP levels and TIg E levels comparison,if variance,uses the independent sample t test,conversely,uses t’-test, The group between different genotypes of ECP level and level of TIg E compared by using analysis of variance,ECP levels between different genotypes and TIg E two comparison using the LSD test.Results: â‘ Asthma group and the control group of sexdifference had no statistical significance(P>0.05),age comparative differences had no statistical significance(P>0.05); â‘¡There are three genotypes as AAã€AG and GG in IL-17A-152G/A in both asthma and control groups,The genotype frequency distribution in the two groups were evidently different(P<0.05),there was statistical significance between AA genotype frequency with AG and GG genotype frequency(P=0.012,OR=2.134,95%CI=1.174-3.877),A and G allele comparative difference was statistically significant(P=0.01,OR=1.477,95%CI=1.097-1.988);â‘¢There are three genotypes as TTã€TC and CC in IL-17 F 7488T/C in both asthma and control groups,The genotype frequency distribution in the two groups were evidently different(P<0.05),there was statistical significance between TT genotype frequency with TC and CC genotype frequency(P=0.022,OR=1.648,95%CI=1.074-2.529),T and C allele comparative difference was statistically significant(P=0.022,OR=1.534,95%CI=1.062-2.216); â‘£In the two groups,serum TIg E level difference was statistically significant(P<0.05);⑤At IL-17A-152G/A sites,in the asthma group,the comparison of TIg E level between different genotype(P<0.05),TIg E levels of AA genotype was higher than AG and GG,and in the control group,there was no difference;â‘¥At IL-17 F 7488T/C sites of the two group,there were no difference in three genotype of TIg E; ⑦Asthma group serum ECP levels was higher than the control group,the difference was statistically significant(P<0.05);â‘§In the IL-17A-152G/A there has no difference between the ECP levels with AAã€AG and GG genotype;⑨In the IL-17 F 7488T/C there has no difference between the ECP levels with TTã€TC and CC genotype. Conclusion: â‘ IL-17A-152G/A locus is one of the important candidate genes of Guiyang region of asthma; The A allele on-152 locus of IL-17 A is associated with children asthma; The TIg E levels with A allele at IL-17A-152G/A locus was higher than without it in the asthma group; â‘¡IL-17 F 7488T/C locus is one of the important candidate genes of Guiyang region of asthma; The C allele of IL-17 F 7488T/C locus has a weak protection role to asthma; The SNP of IL-17 F 7488T/C locus can not influence the level of serum TIg E; â‘¢Serum ECP levels may become the detection index of asthma; There was no correlation between serum ECP levels and the SNPs of IL-17A-152G/A and IL-17 F 7488T/C. |
