ObjectiveTo observe the impact of cluster needling of scalp combined with rehabilitation on the expression of vascular endothelial growth factor and microvessel density in rats after cerebral ischemia.MethodsTo divide 120 rats into five groups randomly:sham operation group,model group,CNSP group,rehabilitation group and CNSP combined with rehabilitation group.Each group were divided into four subgroups in accordance with the 3rd,7th,14th,21 st(n=6).Establish a permanent focal cerebral ischemia model..The rats in sham operation group and model group had no any intervention,the rats in rehabilitation group were trained by treadmill training,the rats in CNSP group were treated by CNSP,and the rats in the CNSP combined with rehabilitation group treated by CNSP and treadmill training at the same time.Neurological impairment evaluated by mNSS and immunohistochemistry staining was used to observ the expression of VEGF in cortical ischemia area at each time point of the rats in each group and weidner method was used to observe the microvessel density in rats after modeling.Results(1)Scores of' mNSS:Sham group have no significant neurological deficit at each time point.After cerebral ischemia 7d,14d compared with the model group,rats in rehabilitation group,CNSP group and the rats in the CNSP combined with rehabilitation group mNSS scores decreased clearly(P<0.05).Compared with the CNSP group and rehabilitation group,CNSP combined with rehabilitation group of neurological severity score was significantly lower(P<0.05).After cerebral ischemia 21d,to compare with rehabilitation group,CNSP group,CNSP combined with rehabilitation group was no significantly statistical difference.(P>0.05)(2)After cerebral ischemia 3d,to compare with the sham group rats in model group have significant differences(P<0.05);to compare with the model group,the number of VEGF positive cells increased clearly in rehabilitation group,CNSP group,CNSP combined with rehabilitation group(P<0.05).After cerebral ischemia 7d,compared with model group,rehabilitation group,CNSP group,CNSP combined with rehabilitation group have significantly statistical difference(P<0.05).CNSP combined with rehabilitation group exceed than rehabilitation group and CNSP group(P<0.05).After cerebral ischemia 14d,compared with model group,rehabilitation group,CNSP group,CNSP combined with rehabilitation group have significantly statistical difference(P<0.05).CNSP combined with rehabilitation group exceed than rehabilitation group and CNSP group.Among the rehabilitation group,CNSP group,CNSP combined with rehabilitation group shows statistically significant(P<0.05).After cerebral ischemia 21 d,the expression of VEGF positive cells in CNSP combined with rehabilitation group were superior to the others(P<0.05).(3)After cerebral ischemia 7d,to compared with sham group the microvessel density of the model group increase obviously(P<0.05).After ischemia 14d,compared with model group,rehabilitation group,CNSP group,CNSP combined with rehabilitation group micro vessel density increased sharply(P<0.05).After cerebral ischemia 21 d.to compare with model group,rehabilitation group,CNSP group,CNSP combined with rehabilitation group have significantly statistical difference(P<0.05).Conclusion1.The method of CNSP combined with rehabilitation can promote the neurological rehabilitation of rats after focal cerebral ischemia.2.The method of CNSP combined with rehabilitation may promote the expression of VEGF in the area of rats after focal cerebral ischemia,which accelerate the process of vascular regeneration. |
