ObjectiveTo explore the effects of cluster needling of scalp acupuncture combined with rehabilitation (Tang’s Approach) on forelimb motor function and expression of synaptophysin and growth-associated protein (GAP-43) in rats after focal cerebral ischemia. And assessment of the unilateral forelimb lesions which was to be involed in the paw reach and grasp motor functional of focal cortical ischaemic lesions in the rat. We observed functional mechanism of cluster needling of scalp acupuncture combined with rehabilitation (Tang’s Approach) on cerebral ischemia in rats.MethodsEndothelin-1(ET-1) was used to induce focal cerebral ischemia model in rats with forelimb dysfunction. The rats were randomly divided into5groups: sham operation group, model group, rehabilitation group, acupuncture group, and acupuncture with rehabilitation group, each group was divided into4subgroups by3d,7d,14d, and21d. and14days before the start of the experiment began forelimbs of rats paw reach pre-training ability, every morning (fasting) one of the training, each15min, so that rat forelimb motor ability to capture food, post-lesion day(D) received the cluster needle of scalp point with forelimb motor ability to capture food in the acupuncture with rehabilitation group,forelimb motor ability to capture food in the rehabilitation group, and the cluster needle of scalp point only were punctured and stimulated in the acupuncture group. But the model group and sham operation group had no intervention.and forelimbs modified method of assessing the rate paw reach test, for postoperative3d,7d,14d and21d between each group and the comparison group observation of the rats at the corresponding time point killed, immuno-histochemical technique was used to observe the expression of synaptophysin and GAP-43in cortical ischaemic lesions of the rats in each group at different time points. Results1.The achievement ratio of the rats that gripped the seeds:before surgery, the rats through pre-training achievement ratio of the rats that gripped the seeds in each group at different time points was no significant difference(P>0.05). and3days after surgery, compared with achievement ratio of rats that gripped the seeds in model group, no difference in the rehabilitation group, acupuncture group(P>0.05), and Tang’s Approach group difference (P<0.05).7days after surgery, compared with model group, all groups were different(P<0.05).14th,21th day after surgery, in addition to the sham operation group, compared with model group, achievement ratio of of the rats gripped the seeds in each group were different(P<0.05). in descending order the order of Tang’s Approach group> rehabilitation group> acupuncture group> model group.2. The expression of SYN:3th,7th day after surgery, each group SYN optical density higher than model group, in descending order the order of Tang’s Approach group>acupuncture group>rehabilitation group>model group.14th,21th day after surgery, in addition to the sham operation group, groups SYN optical density gradually increased, compared with model group were different(P<0.05). in descending order the order of Tang’s Approach group> rehabilitation group> acupuncture group> model group, compared with the rehabilitation group, acupuncture group, model group, SYN optical density of Tang’s Approach group were darker color differences(P<0.05).3. The expression of GAP-43:3day after surgery, in addition to the sham operation group and model group is different(P<0.05), the other groups there was no difference compared with model group(P>0.05).7day after surgery, in addition to the sham operation group,each group of GAP-43positive expression rose to a peak, in descending order the order of Tang’s Approach group> acupuncture group> rehabilitation group> model group.14day after surgery, in addition to the sham operation group, positive expression of GAP-43of other groups were downward trend.21day after surgery, compared with model group, rehabilitation group, acupuncture group, and Tang’s Approach group were different(P<0.05), in descending order the order of Tang’s Approach group> rehabilitation group> acupuncture group> model group, each group with Tang’s Approach group compared with were difference(P<0.05).Conclusion.1.Tang’s Approach can improve the achievement ratio of the focal cerebral ischemia in rats,the promotion of the ipsilateral forelimb grasp, grip, take the recovery of behavioral function,and the Tang’s Approach is better than simply the Rehabilitation and the acupuncture.2.Tang’s Approach could promote expresson of SYN optical density and GAP-43positeve cell cells.3.Tang’s Approach to improve the mechanism of focal cerebral ischemia ipsilateral forelimb motor function may be related to the promotion expression of SYN and GAP-43in surrounding area of cerebral ischemia.
