Part I Determination of exposome of 108 Pesticide in Human Blood SerumA large amount of pesticides are used globally, and the effects of pesticides on human health have aroused attention. With the emergence of the concept of exposome, the determination of exposome makes us more fully understand the relationship between pesticide exposure and human health. However, the methodology focusing on the determination of various pesticides in human blood serum is still scarce, and the exposure level of various pesticides in humans remains largely unknown. We have developed a time-efficient and high-throughput method for the quantification of exposome of 108 pesticides in human blood serum simultaneously using gas chromatography/tandem mass spectrometry. This method is comprised of solid-phase extraction with a small blood serum sample volume(200 ?L)and gas chromatographic analysis with triple quadrupole mass spectrometry.Among the 108 pesticides, limits of detection ranged from 0.01 to 8.00 ng/mL;linearity ranged from 0.02 to 135.00 ng/mL;recoveries ranged from 50 to 150%;intra-day and inter-day relative standard deviations were mostly less than 15%at 2.70 and 27.00 ng/mL.Part II The Expsome of 108 Pesticides in a Non-occupational Population In Jiangsu Province and Safety AssessmentHuman health effects of pesticide exposure have become a social issue. To obtain the pesticide exposome in Jiangsu's population,we determined pesticide exposure profiles using solid-phase extraction and gas chromatography tandem with triple quadrupole mass spectrometry in 200 human blood serum samples from a non-occupational Jiangsu's population.A systematic and comprehensive literature review was carried out to identify the articles investigating pesticide exposure. The developed method showed good analytical performance.Of the 108 pesticides,90 pesticides were found in the blood serum of Jiangsu's population. We found 10497 papers focusing on pesticide exposure. To the best of our knowledge,72 pesticides were firstly found in human blood serum using this new method.By statistical comparison of the blood serum levels of pesticides between this study and other countries,we found several organochlorine pesticides and diazinon were significantly higher in Jiangsu's population.Global health risks related to the pesticide profiling were then assessed. This study not only provides a high-throughput pesticide screening method for future studies of exposome, but also presents the first human data on a number of pesticides and provides a knowledge database for the risk assessment and management of the pesticides.
