Objective:The organochlorines pesticides were called persistent organochlorine pollutants,though it have been disabled for many years, DDTs and HCHs have been always detected in environmental medias and human tissues.Greater attention has been given to the carcinogenic effect of long-term exposed to low doses of organochlorines pesticides.The aim of this research is to investigate exposure level of organochlorines pesticides residues in serum, cancer tissues and non-cancerous tissues from patients with oral squamicous cell carcinoma(OSCC), and then to explore whether the long-term exposed to low doses of organochlorines pesticides can be the risk factor of OSCC.Methods:34new diagnostic OSCC patients and21non-tumor patients hospitalized in the corresponding period from the Hospital Affiliated to Chengde Medical College were enrolled,collected the serum,cancer tissues,non-cancerous tissues and normal oral tissues.Gas chromatography was used to measure the serum and tissues level of organochlorines pesticides residues.The detection ratio difference of organochlorines pesticides in the case and control groups were evaluated by χ2test.The detectable amounts difference of organochlorines pesticides in serum and tissue of cases and control groups were evaluated by Wilcoxon rank sum test and Kruskal-Wallis H test,Nemenyi test. The correlativity analyzing between tissues and surem in the same group were evaluated by Spearman Correlation Analysis,and the difference of organochlorines pesticides detectable amounts were evaluated by Wilcoxon rank sum test.Rseults:1. Comparison of general data between the case and control groups There was no significant difference between the two groups about the age,gender and occupation and so on (P>0.05).2. The detection ratio difference of9organochlorines pesticides in the case and control groups55serum samples were detected,from34patients with OSCC, and21non-tumor patients as controls.8organochlorines pesticides residues including α、β、δ-HCH、P,P’-DDE、P,P’-DDD、O,P-DDT、P,P’-DDT、PCNB could be detected in the case and control groups, but no γ-HCH.In the case, the highest detection ratio was P,P-DDT(79.4%),then β-HCH(70.5%) and P,P’-DDE(50.0%); in the control group, the highest detection ratio was P,P’-DDT(57.1%), then β-HCH(47.6%) and a-HCH(38.0%).According to χ2test,P,P’-DDE and DDTs detection ratio in the case was significantly higher than the control group(P<0.01).85tissue samples were detected, including34cancer tissues and30non-cancerous tissues from34patients with OSCC,21normal oral tissues from non-tumor patients as controls.8organochlorines pesticides residues including α、β、δ-HCH、P,P’-DDE、P,P’-DDD、O,P-DDT、P,P’-DDT、PCNB could be detected in the case and control groups, but no γ-HCH. In the cancer tissues,the highest detection ratio was P,P’-DDE(88.2%),then p-HCH(82.3%) and P,P’-DDT(82.3%).In the non-cancerous tissues, the highest detection ratio was P,P’-DDE(80.0%),then P,P’-DDT(73.3%) and (3-HCH(70.0%). In the normal oral tissues,the highest detection ratio was P,P’-DDT(76.1%),β-HCH(76.1%) and P,P’-DDE(33.3%). χ2test were performed to evaluate the detection ratio difference in case and control groups.The results suggest that in the cancer tissues a-HCH and P,P’-DDE was significantly higher than the normal oral tissues (P<0.05),in the non-cancerous tissues P,P’-DDE was significantly higher than the normal oral tissues (P<0.05),a-HCH in the cancer tissues was significantly higher than the non-cancerous tissues (P<0.05).3. Oganochlorines pesticides residues level in the suremAmong the9Oganochlorines pesticides, β-HCH accounted for the first highest detection amount, P,P’-DDT accounted for the second, and a-HCH accounted for the third, P,P’-DDE for the fifth in the case group. In the control group P,P’-DDT accounted for the first highest detection amount, β-HCH accounted for the second, and a-HCH accounted for the third, P,P’-DDE for the seventh.According to Wilcoxon rank sum test, P,P’-DDE in the cases was significantly higher than the control groups (P<0.05). In the cases the detectaion amount of DDTs was significantly higher than HCHs (P<0.05).4. Oganochlorines pesticides residues level in the tissuesAmong the9Oganochlorines pesticides,(3-HCH accounted for the first highest detection amount, P,P’-DDT accounted for the second, and P,P’-DDE accounted for the third in the cancer tissues and non-cancerous tissues.In the normal oral tissues, β-HCH accounted for the first highest detection amount, P,P’-DDT accounted for the second, and a-HCH accounted for the third, P,P’-DDE for the fourth. Kruskal-Wallis H test and Nemenyi test were performed to evaluate the detection amounts difference in three kinds of tissues groups. The results suggest that a-HCH in the cancer tissues was significantly higher than the non-cancerous tissues(p<0.05),P,P’-DDE in the cancer and non-cancerous tissues were significantly higher than the normal oral tissues(p<0.05). In three kinds of tissues groups, HCHs and DDTs detectaion amounts have no statistically significant difference(p>0.05).5. Compares the residual levels of9organochlorine pesticides between tissues and serum5.1The Correlation of organochlorine pesticides detection amounts between tissues and serumSpearman Correlation Analysis were performed to evaluate the correlation about β-HCH、P,P’-DDE and P,P’-DDT, HCHs and DDTs detection amounts which detection ratio relatively higher than others, between tissues and serum. The results suggest that their have no significant relevance(p>0.05).5.2Comparison between cancer tissues, non-cancerous tissues and serum of the case groupChoose β-HCH、P,P’-DDE and P,P’-DDT,HCHs and DDTs, which detection ratio relatively higher than others,between tissues and serum,compares their detection amounts between cancer tissues and serum of the case group. According to Wilcoxon rank sum test,all the five in cancer tissues and non-cancerous tissues was significantly higher than the serum (P<0.05).5.3Comparison between normal oral tissues and serum of the control groupAccording to Wilcoxon rank sum test,β-HCH、P,P’-DDE and P,P’-DDT,HCHs and DDTs detection amounts have no statistically significant difference between normal oral tissues and serum of the control group(p>0.05).Conclusion:1.8organochlorine pesticide residues can be measured in both serums and tissues in the case and control groups, but no γ-HCH.The residues level of β-HCH、P,P’-DDE and P,P’-DDT was higher than other isomers.2. In the serums, cancer and non-cancerous tissues, the detectaion ratio and amount of P,P’-DDE were significantly higher than the controls. The detectaion amounts of P,P’-DDE in cancer and non-cancerous tissues were significantly higher than its in the serum of the case group,but in normal oral tissues and the serum of the control group its have no statistically significant difference.So P,P’-DDE residuals in OSCC patients may have the relevance with the risk of OSCC.
