Objective:To expore the effects of erythropoietin on IL-4,IL-5,IgE and pathological changes in mice having immediate asthmatic reaction and the influence of related mechanisms.Methods:We choose 54 Balb/c mice,mices of age for six weeks to eight weeks,which is divided into 6 groups according to random grouping method:normal control group,asthma model group,low,middle,high dose erythropoietin group,dexamethasone group.Each group has nine mice.In asthma model group,low,middle,high dose erythropoietin group and dexamefhasone group every 7 days intrapertoneally injection of ovalbumin 20?g(ovalbumin OVA)+ 2 mg aluminum hydroxide,a total of 0.2 ml is gived to the mice and then repeat sensitization.On 22 ed day we make mice asthmatic,with 3%OVA solution atomization inhalation,breathe in 30 min,for 7 days.Before the atomizing inhalation,low dose(500IU/Kg),moderate dose(1000IU/Kg),high dose(2000IU/Kg)of EPO and dexamethasone(1mg/Kg)was respectively injected into enterocoelia in low,middle,high dose erythropoietin group and dexamethasone group.After mice stimulated,there are typical asthma symptoms,such as restlessness,cough,cyanosis,shortness of breath andso on,proving that,asthma model was made successfully.Normal control group have the same method and time of intraperitoneal injection and atomizing inhalation,only use the physiological saline instead of ovalbumin.Detect the levels of IL-4,IL-5 and IgE in serumwith ELISA method.HE staining just after pathological section,and finally we can observe its pathological changes.Results:IL-4,IL-5 and IgE level of serum were increase in asthma model group than that of control group,the comparative diffenence have statistically significant(P 0.05).IL-4,IL-5 and IgE level of serum were significant decrease in low,middle,high dose erythropoietin group and dexamethasone group than that of asthma model group:? IL-4,IL-5 and IgE level of serum were lower in middle,high dose erythropoietin group and dexamethasone group than that of asthma model group,The differences have statistical significances(P<0.05).?IL-4,IL-5 and IgE level of serum were lower in low dose arythropoietin group than that of asthma model group,but the differences don't have statistical significances(P>0.05).We make a comparition among EPO group with different concentrations:?Comparing with the three groups,IL-4,IL-5 and IgE level of serum were significant decrease in middle,high dose erythropoietin group than that of low dose erythropoietin group,the differences have statistical significances(P<0.05).? Make the comparing between the two groups,IL-4,IL-5 and IgE level of serum were significant decrease in high dose erythopoietin group than that of middle dose erythropoietin group,but the differences have not statistical significances(P>0.05).Comparison between EPO groups with different concentrations dexamnethasone group:?IL-4,IL-5 and IgE level of serum were higher in low dose erythropoietin group than that of dexamethasone group,the differences have statistical significances(P<0.05).?IL-4,IL-5 and IgE level of serum were significant higher in middle,high dose erythropoietin group than that of dexamethasone group,but the differences have not statistical significances(P>0.05).(2)After hematoxylin and eosin staining to observe pulmonary histopathologic change by microscopically:The mice in the normal control Bronchial epithelial is intact,non-specific inflammatory response and lung histological morphologic is normal;In the asthma model group results showed that central bronchi and vessels have significant inflammatory cellular infiltration,lumen is stenotic;The low dose erythropoietin group results showed that inflammatory cellular infiltration,lumen stenotic reduce than asthma model group,with the increasing of the concentration of EPO,lung tissue injury was relieved;When Erythropoietin group were Compared with dexamethasone group,we know that lumen stenotic and thickening of smooth muscle are similar,but dexamethasone group have lower inflammatory cellular infiltration.Condusion:EPO can decrease levels of IL-4,IL-5 and IgE in serum and improve lung tissue pathological changes in mice,indicate that erythropoietin have anti-inflammatory effect and definite protecting effect,and the effect were showed dose dependent.
