The Prevention And Treatment Of Tangningtongluo For Hyperglycemia And Complication In Db/db Mice

ObjectiveDiabetes mellitus is a metabolic disorder characterized by hyperglycemia and diabetic complication such as diabetic retinopathy,hepatopathy,angiocardiopathy and nephropathy.Tangningtongluo(TNTL),a traditional MIAO herb,has been proved to be effective on type 2 diabetes mellitus.In the present study,the effects of TNTL on hyperglycemia and diabetic complication in db/db mice were evaluated.Methods1.The present study included two experiments:TNTL pretreatment and TNTL posttreatment.1.1 TNTL pretreatmentDb/db mice(n = 50)aged 6-8 weeks were randomly assigned to 5 groups,1.e.,the model group,the metformin(0.14 g/kg)group,the high dosage of TNTL(1.8 g/kg),the middle dosage of TNTL(0.9 g/kg)and the low dosage of TNTL(0.45 g/kg).The wide-type littermates were used for the control mice.Metformin and TNTL were freshly prepared and intragastric administration daily for 20 weeks.The mice in control group and model group were given the same amount of saline.1.2 TNTL posttreatmentDb/db mice(n = 48)aged 26-28 weeks were randomly assigned to 6 groups,i.e.,the model group,the metformin(0.14 g/kg)group,Jinqijiangtang(2.25g/kg)group,the high dosage of TNTL(1.8 g/kg),the middle dosage of TNTL(0.9 g/kg)and the low dosage of TNTL(0.45 g/kg).The wide-type littermates were used for the control mice.Metformin and TNTL were freshly prepared and intragastric administration daily for 19 weeks.The mice in control group and model group were given the same amount of saline.2.Measurement IndicatorsTo investigate the effects of TNTL on hyperglycemia in db/db mice,we detected the level of fasting glucose,HbA1c,and oral glucose tolerance.To explore the effects of TNTL on lipid metabolism in db/db mice,we determined the level of CHO,TG,HDL and LDL.To explore the effects of TNTL on diabetic complication in db/db mice,the level of microaneurysm were determined by 4Doct fluorescence angiography,the levels of CHO,TG,ALP,CK,GPT,GOT,urine protein,BUN and CRE were detected by respective kits.ResultsTNTL pretreatment and TNTL posttreatment significantly decreased fasting glucose and HbA1c,improved oral glucose tolerance,decreased the levels of CHO,TG,ALP,CK,GPT,GOT,urine protein,BUN and CRE,and reduced microaneurysm.ConclusionTNTL exerted a strong hypoglycemic effect and had preventive effects on diabetic complication in db/db mice.
