Objective:With the expansion of the animal breeding industry and the increase in the number of pet owners,the incidence of animal diabetes is rising year by year.As the most serious complication of diabetic microvascular complications,the incidence of diabetic nephropathy is as high as 30%-40% in recent years.In recent years,a large number of studies have shown that inflammatory response is closely related to the occurrence and development of diabetes,and diabetic nephropathy is considered to be an inflammatory disease.At present,the treatment of diabetes mainly adopts insulin injection,oral hypoglycemic western drugs or traditional Chinese medicine.Although these drugs have certain effects on blood glucose control,they all have certain side effects,and the control effect of diabetes complications is not ideal.Therefore,the search for a hypoglycemic drug that has little harm to the body and can control the occurrence and development of complications has become the focus of researchers in recent years.Taurine as a conditioned essential amino acid in the organism has a variety of biological functions.Our research group and relevant studies at home and abroad have shown that taurine can not only reduce blood glucose and improve diabetes symptoms in diabetic animals,but also inhibit the level of inflammatory cytokines to reduce the damage caused by inflammatory response.It is speculated that taurine may control the development of diabetic nephropathy by inhibiting the inflammatory response.In this study,the diabetic mice model was induced by tail vein injection of STZ,and the preventive effect of taurine on diabetic nephropathy in mice was investigated from the perspective of inflammation inhibition,so as to provide theoretical basis for the application of taurine to prevent diabetic nephropathy in animals.Methods:Built by the method of tail vein injection of STZ mice diabetes model,then the mice were randomly divided into 6 groups of blank control group(C group)respectively,the taurine control group(T group),diabetic model group(DM group)and 1% taurine prevention group(DMT1 group),2% taurine prevention group(DMT2 group),3% taurine prevention group(DMT3 group).The clinical manifestations of mice in each group were observed daily,including mental state,food intake and water intake.The body weight and blood glucose levels of each mice were measured at 2W,4W,6W and 8W,serum and kidney samples were collected at 8W,and the kidney weight was weighed;Levels of inflammatory cytokines IL-6,IL-1? and TNF-? were detected in serum and kidney tissues of mice by ELISA;The degree of neutrophil infiltration was determined by detecting MPO activity in renal tissues of mice in each group;The contents of creatinine and urea nitrogen in serum of mice in each group were determined by microplate method;The structures of glomeruli and tubules were observed by HE;The levels of CD68 and MCP-1 in mice kidney tissues were detected by IHC;Protein expression levels of HMGB1,TLR4,NF-?Bp65,My D88,CD68 and MCP-1 in mice kidney were detected by western-blot.To investigate the preventive effect of taurine on diabetic nephropathy and its mechanism based on NF-?B pathway.Results:Behavioral observation of mice in each group.The results showed that compared with DM group,DMT3 group mice were close to C group in food intake,water intake and mental state;By monitoring the blood glucose and body weight of the mice in each group,the results showed that the blood glucose of the DM group was significantly higher than that of the C group(P< 0.001),the body weight of the DM group was significantly lower than that of the C group(P< 0.001),while the blood glucose and body weight of the DMT3 group were significantly lower than that of the DM group(P< 0.001);The results showed that the renal index of DM group was significantly higher than that of C group(P< 0.001),and the difference between DMT3 group and DM group was more significant(P< 0.001);The serum levels of IL-6,IL-1? and TNF-? in the DM group were significantly higher than those in the C group(P< 0.001),while the serum levels of IL-6,IL-1? and TNF-? in the DMT3 group were significantly lower than those in the DM group(P< 0.001).The levels of IL-6,IL-1? and TNF-? in the kidney of mice in the DM group were all higher than those in the C group,while the levels of IL-6,IL-1? and TNF-? in the DMT3 group were all lower than those in the DM group;The MPO activity in the kidney tissues of mice in each group was significantly higher in the DM group than in the C group(P< 0.001),while the MPO activity in the DMT3 group was significantly lower than that in the DM group(P< 0.001);The content of creatinine and urea nitrogen in serum of mice in each group was detected by microplate method.The results showed that the content of creatinine and urea nitrogen in THE DM group was significantly higher than that in the C group(P < 0.001),while the content of creatinine and urea nitrogen in the DMT3 group was significantly lower than that in the DM group(P < 0.001);The morphological structure of glomeruli and tubules of mice in each group was observed by HE.The results showed that the glomeruli and tubules of mice in the DM group had been pathologically damaged compared with those in the C group,while the morphological structure of glomeruli and tubules in the DMT3 group tended to those in the C group;The levels of CD68 and MCP-1 were detected by IHC.The results showed that the levels of CD68 and MCP-1 in DM group were significantly higher than those in C group(P< 0.001),while the levels of CD68 and MCP-1 in DMT3 were significantly lower than those in DM group(P< 0.001);The protein expression levels of HMGB1,TLR4,NF-?Bp65,My D88,CD68 and MCP-1 in kidney tissues were detected by Western-blot.The results showed that the protein expression levels of mice in the DM group were higher than those in the C group at different degrees,while those in the DMT3 group were lower than those in the DM group at different degrees.The results showed that taurine had a positive preventive effect on diabetic nephropathy by inhibiting renal inflammation.
