Amaranth is a vegetable of amaranth family.The thesis studies effects of different Ca2+?Mg2+?Zn2+?Fe2+?Mn2+concentration of on the growth of Amaranth seedlings.Meantime,based on bioinformatics analysis,eight Zn transporter-encoding genes were cloned and their Zn transportation activity was confirmed with yeast mutant system.Experimental results are followed:?1?It was studied of effects of Ca2+?Mg2+?Zn2+?Fe2+?Mn2+at different concentration on root and hypocotyl length,fresh weight and root hair growth.Under both pH 5.7 and 7.2,Ca2+deficiency and Fe2+oversupply inhibited the growth.Other mineral ions had no significant effects on it.?2?Increasing concentration of Fe2+could partially alleviate the inhibitory effect of Ca2+deficiency on the seedlings'growth.Increasing concentration of Ca2+could alleviate inhibitory effect of Cd,but not the high Fe2+on the growth.The growth of Amaranth seedlings was insensitive to the concentration of Zn2+and Zn2+had no effect on the inhibitory effect of high Fe2+.?3?Transcriptome analysis with the seedling's RNA identified eight intact Zn2+transporter encoding genes named as:AmZTP29?AmZIP11?AmZIP2?AmZIP4?AmZIP1.1?AmZIP1.2?AmZIP1.3?AmZIP7.Among them,AmZIP4?AmZIP1.1?AmZIP1.2?AmZIP1.3 and AmZIP7 could complement the growth of the Zn2+transporter-deficit yeast on the low medium. |
