| Gelsemium elegans benth(GEB)has anti-inflammatory,immune-enhancing,improved intestinal morphology,and adjustment of cecal flora.It is often used to promote the growth of livestock and poultry in the breeding industry.The mechanism of GEB growth may be related to the improvement of intestinal morphological structure,regulation of intestinal flora and anti-inflammatory effects,but the study of the effect of GEB on diarrhea has not yet been reported.The previous research of the team showed that in the GEBEE,GEBEW and GEBEA,the effect of promoting GEBEE growth was the best.Therefore,this experiment is to observe the GEBEE to anti-diarrhea effect of the substance and the preliminary discussion on the mechanism of anti-diarrhea.1.Extracting GEBEE from GEB,using acid dyes to determine the alkaloid content in the GEBEE;using acute toxicity test to determine the toxicity of the GEBEE to determine the appropriate.The dosage of castor oil and senna-induced mouse diarrhea was established to observe the effect of GEBEE on diarrhea in mice,and the GEBEE on the propulsion of small intestine in mice was determined.The results showed that the alkaloid content in the GEBEE was 4.01%,and the suitable high,medium and low doses of the GEBEE were:30 mg/mL,20 mg/mL,10 mg/mL;The diarrhea index of the model group of castor oil-induced diarrhea was(164.20±23.27)%,and the diarrhea index of mice with high,medium and low doses of GEBEE was(105.22±19.38)%,(108.17±39.35)%,(111.12±26.87)%;The diarrhea index of senna-induced diarrhea model group was(213.00±66.00)%,and the diarrhea index of mice with high,medium and low doses of GEBEE was(87.04±31.36).%,(102.64±41.84)%,(128.20±50.07)%;the small intestine propulsion rate of the mice in the diarrhea group was(75.88±8.89)%,and the small intestine propulsion rate of the high,middle and low dose group of the GEBEE was(58.58±),7.23)%,(64.01±4.68)%,(69.23±8.69)%.The experimental results showed that the GEBEE from all the dose groups could effectively treat diarrhea and significantly inhibit the intestine propulsion in mice.Among them,the high dose group had significant effect(P<0.05).2.Using BL-420E bio-functional experimental system to observe the effect of GEBEE on spontaneous contraction of isolated intestinal muscle and sputum contraction induced by ACh,BaCl2 and HA,and observe the IP3 receptor blocker HP.The sarcoplasmic reticulum Ryanodine receptor blocker RR and the nitric oxide synthase inhibitor L-NAME acted as the GEBEE,and recorded the intestinal muscle motion waveform,average tension and contraction frequency.The results showed that the GEBEE inhibited the frequency and tension of spontaneous contraction of rabbit small intestine smooth muscle in a concentration-dependent manner;it inhibited the intestinal contraction caused by ACh,BaCl2 and HA to different degrees,After treatment with L-NAME,HP,RR,L-NAME significantly enhanced the inhibitory effect of GEBEE on the isolated intestinal contraction,while HP and RR had no significant effect.The results showed that the ethanol extract of scutellaria can significantly inhibit the frequency and tension of rabbit small intestinal smooth muscle contraction,and its mechanism may be related to blocking M receptor and H1 receptor,increasing nitric oxide(NO)concentration,and inhibiting IP3 receptor.Mediated intracellular calcium release was associated,but not related to intracellular calcium release by the sarcoplasmic reticulum Ryanodine receptor pathway.3.Established a model of acetic acid inflammation in mice,and injected Evans blue into the tail vein to determine the effect of GEBEE on the content of Evans blue exudate in the peritoneal cavity of mice.The results showed that the high and medium dose groups of the GEBEE were low.The amount of Evans blue exudation in the rat abdominal cavity was significantly decreased(P<0.05);4.Establish a rat model of senna-induced diarrhea.The levels of gas trointestinal hormones(GAS,VIP,MTL)and inflammatory factors(IL-1β,IL-10,TNF-α)in serum were determined by ELISA.The high,medium and low dose groups of GEBEE have elevated MTL and GAS levels,decreased VIP levels,and increased inflammatory factors IL-1βand TNF-α,as well as the reduction of anti-inflammatory factor IL-10.The degree of recovery,especially in the high-dose group works best. |
