| Pump/Sodium hydrogen exchange NHE is a kind of important acrossthe plasma membrane of the ions carrier that exists in eukaryotic cell membranes, thathas a variety of functions, the main function is to regulation the PH of intracellularã€control the size of cells and is responsible for the electroneutrality Sodium/hydrogenion transport, multiple functions such as regulating cell cycle and cell proliferation.Change the activity of NHEs will cause absorption disorder of Na+and diarrhea. Byregulating the activity of NHEs inhibit diarrhea occur will be a new direction toprevention and treatment of diarrhea. The relationship between NHEs gene andintestine has been little reported. The acute diarrheal model was generated using LPSon the basis of previous studies, assessed by observing metal state,intestinalmorphology and peristalsis. The NHEs gene expression of different parts of intestinewas quantified by Q-RT-PCR.We also studied its differential expression patternsbetween control and LPS-injected group.The main results were listed below:diarrhea at different levels, dispiritedness and reduced feed intake occured atdifferent time points(1h,8h) after LPS inoculation compared to control group.Camerawas used to observe the changes of intestine. Among LPS treated mouse, the intestinemoved violently. Mesenteric vein was apparently congested and over timepost-inoculation it looked atropurpureus. The intestinal wall was swelled, and thechange had aggravated compared with the previous. The morphological changes ofintestine were as follows:at1h after injecting LPS, mucosa and villi were slightlydisorganized, A few villous tips were denuded. Degeneration and necrosis of mucosawere not founded,The subepithelial space were expanded.There were slightly lesiongland and congestive capillary. At8h after LPS-injected, villi had significant edema,Mucosa and villi were lined up in disorder with the development of massivedenudation and necrosis, we also found gland was damaged and capillary wasobviously congested, Inflammatory cell concerned infitration appeared on infarct zone.All above pointed out that LPS successfully induced diarrhea, and the function ofintestinal secretion increased in the experimental group.Real-time quantitative PCR was used to quantitatively examine expression levelsof the NHEs gene family. The results show that NHE2, NHE3, and NHE8were predominantly seen in duodenum, jejunum, ileum and ascending colon;NHE2abundance was found in decreasing order: jejunum>duodenum=colonileum;NHE3abundance was found in decreasing order: colon> duodenum=jejunum> ileum;There are no significant differences in the expression levels of NHE8among differentcolonic segmentsï¼›NHE2gene expression is up-regulation in duodenal after LPSinoculation1h,but at8h NHE2gene expression is de-rating. LPS can induce NHE2gene expression de-rating in jejunum, but in jejunum up-regulation, which depend onthe duration. LPS can induce NHE3gene expression up-regulation in the duodenum,jejunum, ileum and colon, which depend on the duration, the expression levels ofNHE2are significant differences in the duodenum, jejunum, ileum and colon at8h.NHE8gene expression is up-regulation in duodenal after LPS inoculation1h,but at8h NHE8gene expression is de-rating; LPS can induce NHE8gene expressionup-regulation in jejunum, ileum,but de-rating in colon, which depend on theduration.The study suggests that diarrhea induced by LPS is closely related with thechange of the transcriptional level of NHEs gene and the effect of different segmentsof intestine on the diarrhea is different. |
