Zebrafish have many unique characteristics such as small size,external fertilization,rapid development,and extreme sensitivity to drug administration,which is an ideal model for screening and evaluating drugs.Liver injury and inflammatory response are diseases with a high incidence in disease research.Research on drugs for treating liver injury and anti-inflammatory has great research prospects.In this paper,thioacetamide?TAA?-induced zebra liver injury model and lipopolysaccharide?LPS?-induced zebrafish inflammation model were used to evaluate lychee ethanol extract?SPEE?,baicalein?BA?,quercetin?Que?and The antioxidant,anti-inflammatory and liver-protective effects of chlorogenic acid?CGA?.Gas chromatography-mass spectrometry?GC-MS?quantitative analysis methods,enzyme activity determination,immunofluorescence staining,Western blot,agarose gel electrophoresis,PCR and Q-PCR were used.The main findings are as follows:1.Lychee grass has antioxidant and anti-inflammatory effects on zebrafish embryos stimulated by LPS.Second,SPEE can significantly reduce TAA-induced hepatic vacuoles,lipid accumulation,and liver fibrosis in adult zebrafish.At the same time,the contents of NO and lipid metabolism-related genes ApoE and PPAR-?were significantly reduced after SPEE exposure.Mechanistically,SPEE can reduce hepatocyte apoptosis and promote the transfer of transcription factor?Nrf2?from the nucleus to the cytoplasm in the TAA-induced model.Finally,SPEE can regulate various inflammatory cytokines and innate immunity-related genes under H2O2stimulation.2.Baicalin improves TAA-induced liver developmental toxicity in zebrafish.It was found that baicalein reduced the level of oxidative stress by enzyme activity measurement,and baicalin was found to have anti-inflammatory effects by inflammatory factor detection.Western Blot results showed that the expression of apoptotic proteins increased and the expression of anti-apoptotic proteins decreased.PCNA staining was found to reduce the number of proliferating cells.Mechanistically,baicalein promotes PPAR?activation by increasing p38 and ERK1/2,thereby regulating liver development.3.Through body length,liver,pancreas and other sizes,it was found that quercetin can effectively promote zebrafish development.At the same time,through enzyme activity measurement and inflammation gene detection,this study found that quercetin has antioxidant and anti-inflammatory effects.In studying the mechanism of quercetin's anti-developmental delay in zebrafish developmental delay,it was proved that quercetin can promote the development of zebrafish liver by regulating the Wnt signaling pathway.4.Chlorogenic acid can effectively promote zebrafish liver development and has antioxidant and anti-inflammatory effects.When studying the molecular mechanism of chlorogenic acid in the treatment of liver dysplasia,it was found that chlorogenic acid can treat TAA-induced zebrafish liver dysplasia by regulating the Wnt signaling pathway.The results showed that lychee,baicalein,quercetin and chlorogenic acid had antioxidant,anti-inflammatory and liver-protective effects.Provide research basis for further opening up new drugs and health foods.
