Intestinal inflammation in animals is very harmful to animals,affecting the digestion of food and the absorption of nutrients.There will be clinical symptoms such as loose stools,vomiting,elevated body temperature,and depression.At present,most of the treatment of animal intestinal inflammation is antibiotic treatment,which is prone to drug resistance,so it is necessary to develop other drugs to replace antibiotics for animal treatment.Quercetin,as a natural flavonoid,exists in a variety of plants.This study used lipopolysaccharide(LPS)to induce a mouse enteritis model to study the protective effect of quercetin on enteritis.To provide a new theoretical basis for quercetin in the treatment of LPS-stimulated enteritis.Research methods: The relative survival rate of mouse small intestinal epithelial cells induced by different concentrations of quercetin LPS was measured by CCK-8.6-8 weeks old KM male mice were randomly divided into control group,control group,high-dose group,middle-dose group,and low-dose group,with 9 mice in each group in total.The body weight of the mice was recorded by weighing,and the body weight of the mice before administration was measured before each administration of quercetin,and the mice were administered intragastrically for one week.The control group and the control group were administered with0.9% sodium chloride.Stomach.Two hours after dosing on the last day,5 mg/kg of LPS was administered by gavage,and blood and small intestinal tissue were sampled 22 h.The changes of different concentrations of quercetin on LPS-induced mice were observed by the body weight and blood cell count of the mice.The damage of the tissue was observed by the pathological changes of the small intestinal tissue of the mouse intestinal tissue,the changes of TNF-α,IL-1β,1L-6,IL-12,IL-18 inflammatory factors were measured by ELISA,and the changes of NLRP3,IL-18 and IL-18 were measured by q RT-PCR.Caspase-1,ASC and NF-κB p65 m RNA levels.Test results: In the in vitro study of quercetin,quercetin can also improve the survival rate of LPS-induced mouse intestinal epithelial cells.Quercetin In in vivo studies,quercetin reduced weight loss in mice,lowered levels of inflammation in the mice’s blood,and reduced intestinal damage in the small intestine.ELISA results showed that compared with the LPS group,quercetin reduced the expression levels of TNF-α,IL-1β,IL-6,IL-12,and IL-18 in the intestinal tract of mice.Quercetin can effectively improve intestinal inflammation caused by LPS by regulating NLRP3 signaling pathway and NF-κB signaling pathway,reduce cell pyroptosis,and protect tissue from damage.Conclusion: Quercetin can protect MODE-K cells and reduce the death induced by LPS stimulation.Quercetin has protective effect on LPS-induced enteritis in mice... |
