Cassava is a root crop of Euphorbiaceae,which not only provides a source of food for nearly 1 billion people,but also provides raw materials for bioenergy.The quality and yield of cassava are affected by the process of plastid division.Therefore,the study of plastid division genes can provide new ideas for cassava variety improvement.Chloroplasts are chlorophyll-containing plastids,which are the sites of photosynthesis in green plants.Its main proliferation mode is binary fission,and one of the main division factors is the evolutionarily conserved Dynamin protein-related protein DRP5B(ARC5).ARC5 belongs to the Dynamin protein superfamily,which plays a key role in a variety of cellular processes,such as vesicle division,organelle fusion and fission,and cytokinesis.Because the Dynamin family and its member ARC5 gene have not been reported in cassava,and the importance of cassava as a "main crop" and its economic potential,its genome has been sequenced,so the cassava genome can be used to systematically study the cassava Dynamin family.Meanwhile,the function of ARC5 gene in cassava was analyzed to explore its importance in cassava plastid division.This study,the Dynamin family in cassava was analyzed systematically and comprehensively by bioinformatics.qRT-PCR was used to analyze the specific expression of cassava Dynamin family member ARC5 in leaves and roots at different developmental stages and to determine the response of ARC5 to abiotic stress.Optimization of cassava seedling hardening technology.Instantaneous Protoplast Expression Experiment to Explore the Localization of ARC5 in Cassava.Construction of overexpression vector and genetic transformation to obtain cassava ARC5 overexpression plants,in order to lay the foundation for subsequent research.The main research results are as follows:1?23 members of cassava Dynamin family were identified and divided into five groups.ARC5(MeDynamin23)belongs to group V.It contains four domains: GTPase domain,MID,GTPase effect domain and PH domain.2?Most MeDynamins were located at the top or end of the chromosome.3?Fragment duplication could be the main replication mode of Dynamin family amplification.ARC5 was highly conservative and had stable genetic characteristics.4?Most members of cassava Dynamin family were expressed in stem apical meristem and embryogenesis,while ARC5 was mainly expressed in leaves.5?ACR5 is closely and positively related to plant growth.High concentration of MeJA treatment led to to the cassava defense mechanism to play its role in advance.salt stress inhibited the expression of ARC5.The trend of ARC5 expression under 20% PEG treatment was similar to that in ABA group.6?Using the optimization method of this study,the survival rate of cassava seedlings was 95%,and the survival rate after transplanting to the field was 100%.7 ? ARC5 are located in chloroplasts.In genetic transformation,the cassava transformation material with infection time of 15 min had the best growth condition when the inoculation solution OD600: 0.3-0.4.
