In this paper,150 persimmon germplasm resources were used as materials to obtain phenotypic traits related to phenotypic trait analysis and cell analysis,which laid a foundation for phenotypic traits;transcriptome sequencing,amplification and analysis of candidate genes were obtained.The genotype and gene association analysis obtained the relevant marker sites,which laid the foundation for molecular marker-assisted breeding of persimmon traits.The relevant research results are as follows:1.Evaluation of Germplasm Resources:150 persimmon germplasm resources of persimmon germplasm resources include 56 phenotypic traits in fruit,seed,pith,pedicel,sepal and fruit handle,and the phenotypic traits are rich in variation.The phenotypic traits of the fruit reflect the size,shape,physical and chemical properties and fruit color.The average seed shape index is 1.81.All the persimmon fruit pith are solid,the pedicels are round and square,with convex,flat and hollow.In the depression,the fruit handle is gradually thickened from the end away the another.2.Diversity analysis:R-type clustering of persimmon germplasm resources can divide 56phenotypic indicators into six groups.Correlation analysis and R-type clustering between indicators indicate size traits and shape indicators of persimmon germplasm resources.There are correlations.Principal component analysis extracted the first eight indicators and explained82.459%of the variation.Q-type clustering can cluster 150 persimmon germplasm resources into 3 categories,each with its corresponding fruit size and shape characteristics.3.Cell analysis:Cell analysis of persimmon fruit at maturity included 34 indicators,28were moderately mutated,and 6 were forced mutations.The average epidermal cell thickness and layer number of persimmon fruit were 87.39 um and 4.1 layers respectively,and the average thickness of peel was 624.69 um.The tannin cells in the cross section were mostly round and the morphological characteristics of tannin cells were determined by longitudinal slitting.The transverse and longitudinal sections of the parenchyma cells are mainly circular,and the number of vascular bundles per unit transverse section is 1/mm~2.Correlation analysis showed that there was a significant positive correlation between cell thickness and cell layer number.The more the number of vascular bundles in the pulp,the more the number of tannin cells.The morphology of tannin cells is divided into polymerization and dispersion.The persimmon tannin cells are small and scattered,and the persimmon tannin cells are large and dense.4.Analysis of related astrigent traits:The average content of soluble tannin in persimmon germplasm resources was 14.61 g/kg.Correlation analysis between soluble tannins content and persimmon phenotypes and cells called significant correlation as astrigent traits.The soluble tannins content were negatively correlated with the size of persimmon fruit,and positively correlated with the content of soluble solids and the number of tannin cells in the area of transverse and longitudinal cuts.In order to quantify the fruit size and shape indicators,the principal component analysis(PCA)of persimmon fruit,seed and pith size and shape indicators related to soluble tannin content was obtained.FAC1 and FAC2 were used to indicate the size and shape of the fruit.Therefore,the indicators of astrigent traits of persimmon fruit were six indicators:including size and shape indicators(FAC1 and FAC2),soluble tannin content(STC),fruit soluble solids content(FSSC),number of tannin cells per unit area of flesh area transverse section(NTC)and longitudinal section(NTF).5.Association analysis:In the transcriptome correlation analysis,150 persimmon germplasm resources were divided into 3 groups,and 6 astrigent traits were obtained by association analysis to obtain 32 SNP loci,and the contribution rate was between 21.41%and32.34%.DKsnp234075,DKsnp90928 and DKsnp482415 are associated with plant resistance proteins.A total of 34 polymorphic loci were found in Dk PA1 gene,and 9 SNP polymorphisms were obtained by association with astrigent traits.Most of the polymorphic loci of astrigent traits were located in intron and exon 2,SNP position 287,874,880 and 994can be subsequently used as molecular marker sites for the astrigent traits.In summary,the fruit phenotypic traits of persimmon germplasm resources in Lanxi City,Zhejiang Province are rich in variation,and the phenotypic variation characteristics of persimmon germplasm resources are obtained.The correlation analysis showed the size and shape of persimmon fruit germplasm resources.There are correlations between the indicators.Cell analysis showed that the resources of persimmon germplasm were rich in cell variability,showing the transverse and longitudinal tannins,parenchyma cell size and morphological characteristics.Correlation analysis of soluble tannins with phenotypic traits and cell traits got six astrigent traits.Transcriptome association analysis screened three SNP loci related to plant resistance proteins,and the association analysis of candidate genes yielded four molecular marker loci that could be subsequently used as astrigent traits.
