Diospyros kaki cv. Youhou belongs to Pollination-constant and non-astringent which was introduced from Japanese. It not only grow well in local area, but adapt extensively, resist harm strongly, fruit early,yield highly and tasted good. In this paper,axillary bud of Diospyros kaki cv. Youhou and seeds of Diospyros lotus L. were used for ex-vivo, cultivation of seedling rootstock as well as shoots-grafting in greenhouse. In this paper, axillary bud of Diospyros kaki cv. Youhou and seeds of Diospyros lotus L. were used for explants for tissue culture, cultivation of seedling rootstock as well as the technology of micrografting in greenhouse. It have studied the disinfecting reagents and processing time of explants, medium of initiating and shoot proliferation culture,and induction of adventitious roots, this results established the in vitro propagation system of Diospyros kaki cv. Youhou and the technology of micrografting in greenhouse.The main result obtained in this study were as follows:(1) Explants selection and sterilization in tissue culture for Diospyros kaki cv. Youhou.The results showed that taking axillary buds with skin of annual shoot as explants, sterilization effect was best, and the explants fast grew. The best process of sterilization for axillary buds with skin was that the explants were immersed in alcohol (75%) for 30 seconds, and then sterilized in HgCl2 (0.1%) for 15 minutes. June would be the best time to collect axillary bud with skin, and pollution rate and the mortality rate of the inoculated explants were significant lower than those in the other months.(2) Established the in vitro propagation system of Diospyros kaki cv. YouhouThe results showed that the fittest medium for germination of stem axillary bud was (1/2 N) MS+1.0 mg.L-1 Zeatin+0.1 mg.L-1IAA.generated shoot were cut as several cluster or single plant, then transferred on (1/2 N) MS+1.0 mg.L-1 ZT+2.0 mg.L-12ip+0.1 mg.L-1IAA, the germination rate could up to 77.8%. when the shoot reached more than 3 cm in length,transferred it in the best rooting medium, the rooting ratio can reached at 73.3% in 1/2 MS(1/2 N)+0.75 mg.L-1IBA after 7 d of darkness culture and 30 d of culture in light. The average root number is 3.6. The survive ratio could reach 85% when they were transplant to outside after be trained.(3) Established the technology of micrografting in greenhouseThe result showed that the seedling basal culm thickness of Diospyros lotus L. can be improved by GA3 treatment with 100 mg.L-1 which can reached at 2.28 cm;the result also showed that the seedling basal culm thickness of Diospyros lotus L. can be improved by silver paper treatment which reached at 1.91 mm, increased 0.44mm campared with controls; PHtohormone treatment can help the recovery speed of micrografting seedlings, the result showed that grafting survival peecentage could be raised by GA3 treatment with 100 mg.L-1.(4) Testing the dynamic changes in pHysiological index of in-vitro micrografting seedlings in the grafting and recovering process.The result showed that the POD and SOD activity of scion had the same varing trend during revovering process, it presented a trend of’increasing-decreasing-increasing’on thewhole. As on the 21 th-28th days after grafting, it can attained maxium and then do down,however, the varing trend of POD and SOD activity of stock have no great difference;the soluble protein content and the soluble sugar content also have same varing trend during revovering process, it presented a trend of’decreasing-increasing-decreasing’on the whole, it have some difference with stock.(5) Established the vitro propagation system of Diospyros kaki cv.YouhouThe result showed that grafting survival percentage could be raised by ZT treatment with 200 mg.L-1which reached at 82.33%, increased 29% campared with controls.
