Thirty-one imported summer millet herbicide resistant strains and optional millet strains were used for variation,clustering,correlation and double correlation analysis on their agronomic traits.Three waxy and two non-waxy millet cultivars were used to analyze polymorphic locis of waxy gene full sequences,amino acid sequence and the secondary domain by Primer 5.0 and DNA MAN software,the main results were as follows:(1)The genetic diversity indexs of the quality traits of the test millet strains were 0-1.910,among them,ear shape and ear stem are larger,plant color and leaf position are smaller;the coefficient of variation of quantitative traits varied from 8.11%to 20.29%%,among them,grain weight and mu yield are heavier,ear diameter and stem thickness are smaller.(2)Yield was very significantly positive correlated to ear stem posture,panicle shape,single-numbered grain number,ear number,1000-grain weight,ear weight,single ear weight,ear diameter,ear length,ear length,but was very significantly negative correlated to thickness of the stem.(3)The test millet lines were devided into six categories at Euclidean Distance 4.0.The first category included 11 summer millet strains,their yield was 6000kg/hm2,their stems were short and thick.The second category was only 2015137-2 Ⅱ14-1.The third category was six summer millet strains,their stems were short and thick.The fourth category included high yield lines such as 2016025♀-2-4(the descendants of Datong 29 series),medium plant height,yield is above 7935kg/hm2.The fifth category included 5 lines of 2015053Ⅰ-2Ⅰ-5-1(the descendants of the Jigu 13 series),their yield was 7335kg/hm2.The sixth category included two lines,2015(07)-4-10 and 2015137-2Ⅱ-5-1,their yield was 7515kg/hm2.A comprehensive analysis of 8 lines with higher yields were comprehensively evaluated,they were 2015(07)-4-10,2015(07)-4-11,20150531-21-4-2,2015053Ⅰ-2Ⅱ-1-1,2015137-2Ⅱ-5-1,16C227-2-2,2016025♀-2-4 and 2016025♀-2-5.(4)The full sequence lengths of the waxy gene were 4218bp.Compared with reference gene sequences,54 mutation sites were detected among five tested millet cultivars.Of them,conversion,transversion(translocation),insertion and deletion mutations were 24,19,5,and 6,respectively.Compared with reference gene sequences,the numbers of different loci of Gonggu68,Hongnigu,Chigu4,Chigu9 and Xinghuangnian were 36,8,5,5,and 0,respectively.Gonggu 68 includes 5 insertions,6 deletions,12 transversions(translocation)and 13 conversion,AGGTG was inserted in Intron9(2883bp)and ACATTCTTTCAGACTGC was deleted in Intron6(2109bp-2125bp).For sequence alignment of waxy gene between non-waxy and waxy millet strains was concerned,Gonggu68,Xinghuangnian and Hongniangu had 31,5 and 3 polymorphic sites compared by Chigu4,respectively.(5)Seventy amino acid sequence different sites were discovered among test millet cultivars,of them 69 sites existed between Gonggu 68 and the other 4 materials,one site was S(serine)in the 248th amino acid from Xinghuang,but that was N(asparagine)from the other 4 millet strains.Five secondary domains of the waxy amino acid sequences were detected in Gonggu 68,they were low complexity(51-67),Pfam:Glycotransf5(81-341),Pfam:Glycostransf1(385-524),Pfam:Glycotrans14(396-533)and low complexity(580-604).The former four secondary domains also detected in Hongniangu,Xinghuangnian,Chigu4 and Chigu9,but was different from Gonggu68 in amino acid positions,the numbers of different amino acids were 0,1,0,and 12 between Gonggu68 and the other four millet cultivars,respectively. |
