Analysis On Agronomic And Qualities Traits Associated With Waxy Gene Exon4-exon9 Interval RT-PCR In Foxtail Millets

51 millet cultivars from Chinese different regions were used for correlation and cluster analysis on agronomic and quality traits. Four different waxy and no-waxy millets were used for RT-PCR analysis in their waxy gene exon4-exon9 interval. The results were listed as follows.(1) Variation range of agronomic traits were 7.89%-49.60% in tested materials, variation of yield was 94.72 kg/mu-484.44 kg/mu, their variation coefficient was 21.81%. The test materials were divided into 7 groups using class average method at distance 4; cultivar numbers in the first class were the most than other classes, associated with 27 varieties such as Dawugu, the seventh class was K97mix2,with the highest yield. Section and leaf number, panicle coarse, panicle weight.panicle spike grain weight and thousand kernel weight were very significant positive correlation with yield, but panicle coarse and the length of the under panicle section, spike length and spike elastic degree were very significant negative correlation, respectively.(2) The protein content of Ji 0515 was the highest than other cultivars, the amount of Datong 34 was the lowest; vitamin B1 content in Gonggu 68 was 0.68 mg/ 100 g; vitamin B2 content in Jiugu 11 was much more than other varieties with 0.72 mg/100g. Selenium content of Xingu 10 and A2-2258 were more than 30μg/kg, which can be used as selenium-rich millet planting processing and production.Gel consistency of 16 cultivars were more than 120mm.46 materials had middle gelatinization temperature.3 copies was waxy type such as Mozhuannian. Amylose content of Baiminiangu and Gonggu 68 were 0, amylose content of 4 cultivars was greater than 17%,39 copies were low amylose type.(3) Variation ranges of quality traits in selected 51 millet materials were 9.38%-128.57%, variation coefficient of protein content was the minimum, and vitamin B2 was the largest. The tested materials were divided into 9 groups using Minimum distance method at distance 10. Amylose content in the fourth and fifth class was 0. Amylose content in the third class was higher than other classes, associated with 34 cultivars. Vitamin Bl had significant positive correlation with vitamin B2; protein content had very significant negative correlation with amylose content.(4) Exon4-exon9 interval sequence alignment showed that waxy millethomology was 99.12% between Hongniangu (00000537) and Baiminianug, with four single nucleotide polymorphism variations. Homology for Fengtian501 and Chigu 4 was 98.9%, associated with five single nucleotide polymorphisms. The homology in Hongniangu(00000537) and Chigu 4 was 98.23%, the single nucleotide polymorphism loci in the former were T、G、T、G、G、A、C and A, which were in 139th、163th、189th、192th、245th、358h、445th、and 451th bases, respectively, but that in the latter were C、A、G、A、A、C、T and G in the same bases, respectively. Six amino acid variants were detected in amino acids sequence alignment, which were canslated from cDNA sequence of Exon4-exon9 interval in Hongniangu (00000537) and Chigu 4.
