Osteoarthritis(OA)is a common joint disease with a serious impact on normal joint function of animals and even animal welfare.Currently,there are a variety of clinical treatment methods,but each treatment method has its own inapplicability,which requires clinical drugs with little side effects,convenience and economy and suitable for animals.Natural products,in recent years,have gradually become central issues.Baicalein,as a nature products,have anti-inflammatory,antiapoptotic and anti-tumor effects.And it is expected to become a drug applicable to the clinical treatment of OA in animals.This study aimed to investigate curative effects by different dose Baicalein in medial meniscectomy(DMM)induced rats Osteoarthritis comparing with dexamethasone.The screened dose of Baicalein was used to cure Groove mediated Osteoarthritis canines and evaluate the curative effect.In rats vivo experiment,medial meniscectomy induced rats were divided into 6 groups,control group,model group,Baicalein low dose group,Baicalein middle dose group,Baicalein high dose group and dexamethasone group.Each group was randomized to 8 SD rats.Later,rats were injected with drugs at 7 days until 6th and 8th weeks.Serum and articular cartilage were separated to analyze the effects of drugs on IL-1?,TNF-?,MMP-13,ADAMTS-5,COMP,CTX-II protein expression quantity.Finally,Tibia issue slices were to stain safranin O solid green and immunohistochemistry.Results showed that the contents of blood IL-1?,TNF-?,cartilage MMP-13,ADAMTS-5,urine COMP and CTX-II in the baicalein-treated group were significantly or significantly lower than those in the DMM group(P < 0.01 or P < 0.05).Reduced cartilage damage.In canine vivo experiment,4 Groove mediate canines were divided into 2 groups,control group,experimental group.Control group was randomized to 1 canine and experimental group 3 canines.Later,canines were intra-articular injected in drugs at 3th until 8th weeks.Blood,urine and joint fluid were collected from all animals 1 week before,4 weeks after,1,3 and 5 times after and 1 week after the administration.After the drugs delivery totally,blood,urine,joint fluid were separated to test IL-1?,TNF-?,COMP and CTX-II protein expression quantity in different treatment timewindow.Finally,we investigated Baicalein curative effects of canine Osteoarthritis by clinical examination,gait assessment,B-ultrasound and X-ray examination.The results showed that the contents of IL-1?,TNF-?,blood COMP and urine CTX-II in the baicalein administration group were significantly or significantly lower than those in the control group(P < 0.01 or P < 0.05).The pain of the sensitive joints was relieved,the symptoms of abnormal gait and posture were relieved,and the range of motion was enlarged.Experiments show that: radix scutellariae element can reduce proinflammatory factor IL-1? and TNF-? the expression of rats and dogs,reduce cartilage matrix-degrading enzymes related to MMP-13 and ADAMTS-5 protein,collagen type II degradation is abate,improve the clinical symptoms of OA canine osteoarthritis,delay the process of OA.
