Guaiazulenum is a natural hydrocarbon extracted from plants and has a therapeutic effect to promote skin wound healing.But for many years,little research has been done on its therapeutic principles.In order to explore the healing effect of guaiazulenumon skin wounds and its principles,and promote its development and utilization,60 healthy SD male rats weighing 220 ± 20 g were used to prepare 2 diameters on their backs using surgical methods.It is a model of 10 mm total skin defect injury,which is randomly divided into a model control group and a model treatment group.On the day of surgery,the model treatment group applied guaiazulenum ointment to their wounds,and the model control group was applied petroleum jelly ointment,which was continuously administered for 21 days,and the photos were taken on the 0th,3rd,7th,14 th,and 21 st days.At 0,3,7,21 days after surgery,the skin around the wound was taken for HE staining,and the pathological changes were observed with light microscope;Masson stained collagen tissue,and the collagen deposition rate at the wound was measured using Image-Pro software;immune CD-34 was detected by histochemical method,and the microvessel density at the wound was measured by Image-Pro software.ELISA method was used to determine the levels of interleukin IL-1?,myeloperoxidase(MPO),prostaglandin E2(PGE-2)and tumor necrosis factor ?(TNF-?)in wound tissues at different times and groups.RT-QPCR was used to detect the relative expression of transforming growth factors TGF-?1,Smad2,Smad3,vascular endothelial growth factor VEGF,hepatocyte growth factor HGF,epidermal growth factor EGF,and fibroblast growth factor FGF-7,and Western blot to detect TGF-?1,VEGF expression levels.Results: The wound healing of the guaiazulenum treatment group was faster than that of the model control group,and the healing rate of the two groups was significantly different at 7 days(P <0.05).HE staining showed that the wounds in the guaiazulenum treatment group had a larger number of connective tissues at 7 days,and the degree of inflammatory infiltration was weaker.Masson staining showed that the collagen deposition rate of the guaiazulenum treatment group rats was higher than the model at all time points.In the group,the difference was significant at 14 days(p <0.05).CD-34 immunohistochemistry showed that the microvessel density of the wound in the guaiazulenum treatment group was higher than that in the model control group at 3 d and 7 d.The guaiazulenum treatment group was 88 ± 24 and 54 ± 18.19,and the model control group was 68 ±.38.35 and 42.33 ± 9.61,but they were smaller than the model control group at 14 d(p <0.05).At 3 days,the results of the treatment group showed that the IL-1? was 9.78ng/L,which was lower than the model control group at 17.48ng/L(p <0.05);the PGE-2 concentration in the guaiazulenum treatment group was significantly lower at 7.94ng/L and 7.12ng/L at 3 and 7 days,respectively.Compared with 8.68ng/L and 7.21ng/L of the model control group(p <0.05);the concentration of TNF-? in the guaiazulenum treatment group was 19.93ng/m L at 7 d,which was significantly lower than that of the model control group of 26.67 ng/m L(p <0.05).RTQPCR showed that the VEGF m RNA of the treatment group was up-regulated at all time points,and the difference was significant at 3 days(p <0.05),the relative expression was(4.04 ± 0.49):(2.00 ± 0.13);FGF m RNA guaiazulenum treatment group all The time point was up-regulated.The relative expression level was(7.44 ± 0.32)at 14 d:(4.53 ± 1.02),the difference was significant(p <0.05);HGF m RNA in the guaiazulenum treatment group was up-regulated at all time points,and the difference was significant at 3 d(P <0.05),(5.22 ± 0.39):(3.86 ± 0.37);the m RNA level of TGF-?1 in the guaiazulenum treatment group was raised at all time points,and the difference was significant at 7 days,(7.41 ± 0.54):(5.15 ± 0.30)(p <0.05);the downstream factors Smad2 and Smad3 were also up-regulated,and Smad3 was significantly different at 3 d and 7 d.Western blot showed that both VEGF and TGF-?1 expression were up-regulated.Conclusion: Guaiazulenum promotes skin wound healing.It mainly promotes this process by inhibiting the occurrence of inflammation and reducing the expression level of inflammatory factors,and can also increase the expression of some growth factors to a limited extent,thereby promoting collagen synthesis and angiogenesis.
