Ecdysterone,as a hormone which regulating the metabolism in insects,has a series of functions such as regulating immunity,promoting protein synthesis,regulating the proliferation of endothelial cells,promoting the growth of damaged cells and improving the healing of skin wounds.Wound healing is a complex biological process which regulated by repair factors.A variety of biologically active substances play an important role in all stages of wound repair such as growth factors,cytokines,collagen fibers,adhesion molecules and chemokines,and the expression of these repair factors in the wound healing process is closely related to the time of the occurrence of the wound.In this study,Wistar rats anddogs were used as experimental animals to study the specific effects of ecdysterone on skin wound healing.In the experiment,48 male Wistar rats were randomly divided into four groups: model group,dissolving medium group,Yunnan Baiyao group and ecdysterone group.Through the wound healing rate,pathological tissue section observation of wound site,the levels of inflammatory factors(IL-1?,IL-6 and TNF-?)in serum and the content of wound healing related cytokines(VEGF,?SMA and ?FGF)to study the effect of ecdysterone on the overall process of wound healing and the specific effects on the two stages of inflammatory and proliferative phases.Four male dogs were divided into four groups,and the healing effect of ecdysterone was verified by the wound healing rate experiment.Experimental results show:The results of wound healing experiments in rats shown that the average wound healing of the wounds in thedrug-administered group and the positive control group were significantly higher than that in the model group and the dissolving medium group(P<0.01;P<0.05)on the 3rd day after trauma.And then the former wound healing rate was still significantly higher thanthe dissolving medium group(P<0.05).The results of pathological section shown that the amount of inflammatory cells in the pathological sections of the drug-administered group and the positive drug group on the third day was less than that in the model group and the dissolving medium group,and granulation tissue and angiogenesis were already present;on the 7th day,inflammatory cells infiltrated in the drug-administered group and the positive group became less,and the cells were mainly fibroblasts which arranged in an orderly manner.But there were still a large number of inflammatory cells infiltrated in the model group and the dissolving medium group and the number of fibroblasts was reduced,and there were less angiogenesis.After the 14 th day,the epidermis of the drug-administered group and the positive drug group were completely replanted,the skin structure was clear,and the epidermis had keratinization.However,the granulation tissue and the regenerative blood vessels in the model group and the dissolving medium group were not degraded.The arrangement of the fiber cells is not very neat.The results of enzyme-linked immunosorbent assay shown that the levels of inflammatory factors IL-1?,IL-6 and TNF-? in the serum of the mice in the drug-administered group were significantly higher than those in the model group and the dissolving medium group(P<0.01).The results of immunohistochemistry shown that on the third day,the average VEGF cumulative optical density,average ?SMA cumulative optical density and average ?FGF cumulative optical density were significantly higher in the drug-administered group than in the model group and thedissolving medium group(P<0.01).The results of dog skin wound healing experiments shown that the epidermal replantation time of the wounds in the drug-administered group and the positive drug group were significantly shorter than that in the model group and the dissolving medium group.The results suggest that ecdysterone can shorten the inflammatory phase of wound healing and increasing the levels of VEGF,?SMA and ?FGF in the wound site during the proliferative phase to promot the wound healing in rats.In addition,ecdysterone can also promote healing in dogs.
