In recent years,due to the aggravation of urban river pollution,the fish resources have been greatly reduced.In order to restore the ecological environment and the amount of fish resources,the stock enhancement program have become increasingly important.Monitoring and evaluation of the discharge effect need effective catch quantity,and the catch rate of the fish is closely related to the choice of marking method.Alizarin complexone?ALC?is a kind of fluorescent marker substance which can be used for a stock enhancement program of fish.This substance has the advantages of low price and long mark retention time for large-scale artificial stock enhancement program,providing a relatively simple and cheap detection method for the evaluation of release effect.But a certain concentration of alizarin complex immerse tag,have a certain toxicity to fish,the greater the concentration,the death rate is usually higher,and the fish for ALC interspecific differences in tolerance and individual size differences,therefore,this study chooses different conditions of marking,according to the possible response of ALC to fish stress and marking effect,determine the appropriate marking conditions,carry out large-scale marking discharge,and preliminarily evaluate the multiplication and discharge effect of urban watershed of bahe river.The main results are as follows1.Alizarin complex is suitable for Aristicluthys nobilia to fluorescent labeling.According to the results of fluorescent labeling and its stress response,it was found that the appropriate concentration for the fluorescent labeling of Aristicluthys nobilia was 100mg/L and the immersion time was 48h.2.By comparing the marking effects of different otoliths under different fluorescence conditions,it was found that microotoliths were the most suitable type of otoliths for detecting alizarin complex for Aristicluthys nobilia,and the best under blue fluorescence conditions.3.According to the effect of fluorescent labeling different feeding density?20,40,50tails/L?,it is found that feeding density has little influence on the effect of fluorescent labeling.Considering economic cost,labor,marking effect,survival rate and other factors,it is considered that the appropriate feeding density is 50 tails/L.4.Under different concentrations?0,50,100,200mg/L?of alizarin complex markers.The relative expression levels of mRNA of cat,cyp1a,mn-sod,cu/zn-sod,gpx,gr,gst and hsp70 were determined;Changes in the enzyme activities of SOD,CAT,Gpx,GST,GPT and GOT,as well as the content of H2O2 and MDA showed that the stress response of bighead fish labeled with alizarin complex with a concentration less than or equivalent to200mg/L was not serious damage.As time went on,the body returned to a normal state.5.According to the marking effect of alizarin complex,it was found that when the marking concentration of alizarin complex was greater than or equal to 100mg/L,the fluorescent marking of Aristicluthys nobilia microotolith could be preserved for at least 1year.6.According to the growth changes of bighead discharge in Bahe urban watershed,,it is found that the growth condition of bighead is good.
