Penaeus japonicus is an important economic and stock enhancement species in China.The stock of Penaeus japonicus have declined sharply in recent years due to environmental change,habitat decline and overfishing.As an effective method to restore fishery resources,stock enhancement has been widely used in aquatic industry.In this study,microsatellite molecular markers were used to analyze the effects of stock enhancement of P.japonicus in Beibu Gulf,and the genetic effects on wild populations.Genetic diversity and population genetic structure of P.japonicus in the South China Sea were studied by using microsatellite markers.The purpose of this study was to evaluate whether the stock enhancement in Beibu Gulf has influences on the P.japonicus,to explore the genetic characteristics of P.japonicus population in the South China Sea,and to provide theoretical basis and data support for the conservation and rational development and utilization of P.japonicus resources in the future.The main results are as follows:Six microsatellite primers were used to analyze the results of stock enhancement in 2019.143 female parent P.japonicus and 663 captured P.japonicus were analyzed.The results showed that 101 of 663 recaptured P.japonicus individuals could be found their corresponding mothers based on 6 microsatellite loci,which accounted for 15.23% of the total recaptured P.japonicus.The genetic effects of stock enhancement on wild populations of P.japonicus were analyzed based on five microsatellite primers.The results showed that the genetic differences among parent populations,released populations and recaptured populations were weak,and the genetic effects of stock enhancement on natural populations were not significant.In order to detect the genetic diversity and genetic structure of P.japonicus populations in coastal China,10 pairs of microsatellite markers from 143 individuals of 5 populations(ZJ,ST,LS,SW and BH)were analyzed.The results showed that there was a moderate degree of genetic differentiation between Shanwei and Shantou populations,but there was a low degree of genetic differentiation among the remaining populations.According to the result of UPGMA tree constructed by genetic distance,the ZJ,LS and BH populations clustered into one branch,and the ST and SW populations clustered into the other branch.The clustering results were consistent with the morphological variation types.
