| Estrogen is an important reproductive endocrine hormone,which is mainly produced by the ovaries.The natural estrogen in the living organism is mainly in the form of 17-βestradiol(17-βestradio,17-βE2),estrone(Estrone,E)and estriol(Estriol,E3),17-βE2 in three Among them,the biological activity has the highest effect and is the most widely used.Estrogen has a wide range of biological effects and plays an important role in various systems of living organisms.In addition to its role in the reproductive system,the motor system,respiratory system,and nervous system are also regulated by estrogen.In previous studies,there are still many contradictions in the regulation of estrogen on gastrointestinal tract activity,but the classical endocrine pathway can not reasonably explain the existence of this contradiction.Then,what is the way estrogen regulates the electrical activity of gastric antrum?Realized?In this study,the expression and distribution of estrogen nuclear receptor(ERα,ERβ)and muscarinic acetylcholine M2 receptor(M2mAchR)in the antrum wall of females were detected by immunofluorescence double labeling technique,and BL-420F was used.The biological function test system monitors the effect of estrogen single factor on the electrical activity of gastric antrum and the effect of estrogen combined with acetylcholine on the electrical activity of gastric antrum,and constructs related mathematical models to analyze its biological significance and elucidate the estrogen pair.The intrinsic mechanism of the antral electromyography activity index,the main results obtained in this test:1.Estrogen receptor(ERα,ERβ)fluorescent staining is negative compared with ERαand ERβpositive controls on the ovary,and the expression of acetylcholine M2 receptor(M2mAChR)is in the gastric antrum muscle layer and mucosa.Muscle layer,mucosa layer and submucosa were distributed.The muscle layer of gastric antrum and gastric antrum mucosa was bright green,which was strongly positive.In the gastric mucosa layer,the submucosa was dark green and weakly positive.It indicates that estrogen does not have a material basis for direct regulation of the gastrointestinal tract through the endocrine pathway.2.Ovariectomy was performed on rabbits with different doses of estradiol.The activity of antral electromyography was monitored by BL420-F.As the dose of estradiol increased,the activity index of antral electromyography increased first and then decreased.Analysis of variance showed that the difference of antral electromyographic activity index between adjacent groups was significant(p<0.05)or extremely significant(p<0.01).Taking the estradiol dose as the independent variable x,the antral electromyography activity index is the dependent variable y,and the fitting is obtained:y=2.80+5.65×exp{-0.5×[(x–0.159)/0.038]~2}is Gaussian The function(R~2=0.9884,P<0.05)indicates that the regulation of estradiol on gastric antral smooth muscle is based on the intrinsic regulation mechanism of gastric antrum itself,and has a great influence on its internal regulation mechanism.3.Two-factor orthogonal grouping design was used to obtain different doses of estradiol/acetylcholine in rabbits.The data of different doses of estradiol/acetylcholine on the changes of gastric antrum electrical activity were collected by BL420-F bio-functional system..The results showed that under different doses of acetylcholine,the antral electromyography index increased first and then decreased with the increase of estradiol dose,showing a Gaussian function.At different estradiol doses,the antral electromyography index With the increase of acetylcholine dose,it showed a trend of increasing first and then gradual,and its characteristics accorded with a logical function.Therefore,estradiol and acetylcholine doses were used as independent variables,and antral electromyography was the dependent variable.The mathematical model with Gaussian function as template was obtained:z=5.11/{1+exp[-52.84×(y–0.016)]}+[3.82+2.40×exp{-exp[-(y–0.042)/0.008]–(y–0.042)/0.008+1}]×exp{-0.5×[(x–0.098)/0.034]2},a mathematical model with a logic function as a template:z=6.75+6.41×exp{-exp(x-0.16)/0.037}/[1+exp{43.55-7.86×exp[-exp(x-0.23)/0.032]-(x-0.23)/0.032+1}×(y-0.01)}].And through two functional analysis,it is indicated that the regulation of estradiol on the electrical activity of gastric antrum is based on acetylcholine.This study,through morphological study,digital-analog analysis showed that the regulation of estrogen on the electrical activity of gastric antrum is based on the indirect pathway of cholinergic nerve.This conclusion complements the neuro-immune-endocrine theory and opens the estrogen to the body.The new vision of the regulation mechanism of internal organs has important theoretical significance.The mathematical model obtained provides an important reference for the accurate application of estrogen drugs in clinical practice. |
