Platyeladus orientalis is an important tree species for afforestation in China,which has the functions of soil and water conservation,wind protection and sand fixation,and landscaping.There are abundant genetic resources in the ancient population in Qiao mountain where the mausoleum for Huang Di is located.It is of great significance for protection and utilization to explore the genetic diversity of the ancient population and formulate appropriate strategies for the conservation of genetic resources.The old tree planted by Huang Di is located in the courtyard of Xuanyuan temple in the mausoleum.Legend has it that it is planted by Huang Di,and it is the world's oldest cypress,with important scientific and cultural value.However,it is difficult to identify the clones of this old tree using phenotypic characters.We used SRAP molecular markers to analyze the ancient population in Qiao mountain and the old tree planted by Huang Di,explore the genetic diversity of the ancient population,and construct the DNA fingerprint of the old tree.It provides a basis for the genetic resource protection of the ancient population and molecular identification of the old tree and it's clones.The main results are as follows:1.The 22 primers amplified a total of 272 fragments,of which 188 were polymorphic,with a polymorphism rate of 69.12%.The polymorphism rate of primers combination Me3Em15 and Me4Em9 was 100%.The lowest polymorphism rate was primers Me4Em10?52.94%?.Each primer amplified an average of 8 polymorphic DNA fragments.At the similarity coefficient of 0.68,ancient trees can be divided into 5 categories.The old tree planted by Huang Di can be divided into 1 category.The second category includes 17 trees,the third category includes 6 trees,the fourth category includes 4 trees,and the fifth category includes 2 trees.2.Six specific markers were detected including Me4Em9-150,Me4Em9-1600,Me4Em9-1700,Me10Em4-80,Me10Em4-90,and Me10Em4-120.If these six markers were used to identify,the probability of error was 5.4x10-8%,and the correct probability was99.9999999%.This indicated that these six markers could be used as DNA fingerprinting for molecular identification of this old tree.
