Fatty liver is a nutrient metabolic disease that often occurs in perinatal dairy cows.It has a serious impact on its production performance,reproductive performance and immune function.Some human medicine research data show that some of the adipokines are involved in the occurrence and development of fatty liver,which can be used as an independent risk factor for fatty liver,and have a certain predictive effect on the occurrence of fatty liver.However,in the dairy cow production,this research has not yet seen reported.Therefore,the purpose of this study was to investigate the association of partial adipokines with hepatic function indexes and energy metabolism indexes in periparturial fatty liver dairy cows and whether the corresponding adipokines in prenatal serum can be used as a risk assessment indicator for postpartum fatty liver.It lays the foundation for revealing the action mechanism of adipokines in fatty liver disease of dairy cows,and provides a theoretical basis for exploring whether adipokines can be used as an early risk assessment index for fatty liver in dairy cows.This study used a semi-enclosed uniform in a large-scale dairy farm in Sichuan,with a weight of 510±47kg,and a total of 84 Holstein cows in the early perinatal period of 1-4 births.According to the incidence of postpartum fatty liver,28 fatty liver cows were selected as the experimental group,and 36 non-fatty liver cows were used as the control group.The remaining 20 cows abandon Venous blood serum was collected at 21 days prepartum,7 days postpartum,and 14 days postpartum early feeding,and the age,parity,and body condition scores were recorded.The content of some adipokines(leptin,adiponectin,TNF-?,IL-6)in serum was detected by ELISA,and the liver function indexes(AST,ALT,TP,ALB,GLB,A/ G,TBIL,DBIL,IBIL)and energy metabolism indexes(NEFA,BHBA,Glu)were measured by biochemical methods.The effects of leptin,adiponectin,TNF-? and IL-6 in serum of dairy cows during perinatal period on the fatty liver of cows and their correlation with liver function and energy metabolism were analyzed;Using binary logistic regression analysis and ROC analysis to determine prenatal risk assessment index of fatty liver in dairy cows and its critical value and predictive effect.the results are as follows:1 Correlation analysis of partial adipokines and fatty liver in perinatal dairy cows The results showed that:(1)There is a extremely significant positive correlation between leptin and adiponectin in dairy cow(p<0.01)and a significant negative correlation with TNF-?(p<0.05);there was a extremely significant negative correlation between adiponectin and TNF-?(p<0.01).;there was a significant positive correlation between TNF-? and IL-6(p<0.05).It shows that there is a close relationship between some of the adipokines in the serum of perinatal cows(2)There was a extremely significant positive correlation between leptin and Glu,TNF-? and BHBA in cows(p<0.01),and there was a significant positive correlation between TNF-? and NEFA,IL-6 and NEFA(p<0.05).There was a significant negative correlation between leptin and BHBA,adiponectin and NEFA,adiponectin and BHBA,TNF-? and Glu(p<0.05);leptin was extremely significantly negatively correlated with NEFA(p<0.01).It shows that there is a close relationship between partial fat factors and energy metabolism indicators in the serum of perinatal dairy cows.(3)There was a extremely significant negative correlation between leptin and AST in serum(p<0.01),and there was a extremely significant positive correlation between leptin and TP,adiponectin and TP(p<0.01);There was a significant positive correlation between ALB,leptin and A/G,adiponectin and ALB,TNF-? and AST,TNF-? and DBIL,IL-6 and ALT(p<0.05);there was a significant negative correlation between leptin and TBIL,leptin and DBIL,adiponectin and AST,TNF-? and TP,TNF-? and ALB,IL-6 and TP,IL-6 and ALB(p<0.05).It shows that partial serum adipokines in the serum of perinatal cows can significantly affect liver function.2 The risk assessment of partial fatty liver factors in fatty liver in perinatal dairy cows.The results showed that serum leptin,TNF-?,IL-6,AST,NEFA,Glu and body condition scores at the 21 st day prepartum can be used to assess the risk of postpartum fatty liver in dairy cows.The best cut-off values were Serum leptin <27.64ng/mL,TNF-?> 110.81ng/L,IL-6>159.14ng/L,AST>75.48U/L,NEFA>0.434mmol/L,Glu < 2.979mmol/L,BCS> 3.625.Conclusion:(1)Leptin,adiponectin,TNF-? and IL-6 in serum of fatty liver dairy cows are closely related to liver function indexes and energy metabolism indexes,and they have different degrees of correlation.(2)The serum leptin,TNF-?,IL-6,AST,NEFA,Glu and body condition scores at the 21 st day prepartum can be used to assess the risk of postpartum fatty liver in dairy cows.
