| Fatty liver is a metabolic disorder that occurs frequently in dairy cows during the perinatal period,and causes great harm to dairy cows,causing serious economic losses.Therefore,early detection and diagnosis is very important for pastures.Liver biopsy is more harmful to dairy cows and the operation is complicated.The Reid equation is used to diagnose fatty liver.The operation is simple but the calculation is complicated,and it is not suitable for large-scale fatty liver detection.Therefore,there is an urgent need to find a diagnosis method that is simple to operate and easy to judge.This study intends to use the Reid equation to determine the fatty liver of dairy cows,and then screen the relevant serum indicators as biomarkers,and then use the receiver operating characteristic(ROC)curve to evaluate the sensitivity,specificity and accuracy of the relevant indicators in diagnosing fatty liver.Find the best critical value.It hopes to provide a new method for fast,simple and accurate diagnosis of fatty liver in dairy cows.Collect serum samples of 77 Chinese Holstein dairy cows after 7 days calving,determine NEFA,Glu,AST,ALT,ALP,TP,ALB,TG,TCHO,HDL-C,LDL-C,Ins,Gln,TGF-β,Apo B,FGF-21 content,according to the Y value calculated by the Reid equation of AST,Glu and NEFA,cows were divided into fatty liver disease group and normal group,and the correlation between serum indexes and Y value was analyzed,and it was found that NEFA,Glu,AST,ALT,TP,TG,LDL-C,Ins,Gln are significantly correlated with Y value.ROC curve is used to evaluate the sensitivity,specificity and accuracy of these indicators in diagnosing fatty liver,and calculate the best cut-off value.This results showed that it can be used as biomarkers for the diagnosis of fatty liver in perinatal dairy cows when the AUC value of NEFA,AST,ALT and Gln is more than 0.5.The diagnostic sensitivity is 81.8%,72.7%,63.3%,54.5%,respectively,and the specificity is 92.8%,89.4%,84.8%,69.7%,respectively.The best cut-off values for the diagnosis of fatty liver with NEFA,AST,ALT and Gln were 471.74 μmol/L,95.5U/L,46.5U/L,64.6mg/m L,respectively.In order to further verify the accuracy of these biomarkers in diagnosing fatty liver,using the serum NEFA and AST data of 461 cows collected in the laboratory,the Y value calculated by the Reid equation is used as the standard to determine the accuracy of fatty liver.The accuracy of the best cut-off values of NEFA,AST and NEFA + AST for the diagnosis of fatty liver are 89.96%,73.49% and 69.08%,respectively.In summary,NEFA,Glu,AST,ALT,TP,TG,LDL-C,Ins and Gln were evaluated in this study.The best cut-off values of NEFA,AST,ALT and Gln for the diagnosis of fatty liver in dairy cows were 471.74umol/L,95.5U/L,46.5U/L and 64.6ng/m L,respectively.According to the serum concentrations of these four biomarkers,fatty liver can be diagnosed directly and accurately,which is convenient for large-scale use in dairy farms. |
