Baitouweng Tang,as the first choice of traditional medicine for dysentery,is widely used in clinical veterinarian.In this study,according to the characteristic and function of traditional prescription,the deficiency of traditional dosage form was overcome,and Baitouweng Tang was developed into compound Chinese medicine powder.To evaluate the effect of this traditional Chinese medicine,a pre-clinical drug effect evaluation test was carried out,providing useful reference for the development of new veterinary-use dysentery medicine.The main contents and results of this study are as follows: 1 The vitro study on the antibacterial and analgesic and anti-inflammatory activities of compound pulsatilla powder 1.1 Antibacterial activity in vitroThe Minimal inhibitory concentration(MIC)and Minimal bactericidal concentration(MBC)of compound pulsatilla powder against Escherichia coli,Staphylococcus aureus and Salmonella were determined by double dilution method.The results showed that the lowest values of compound pulsatilla powder for MIC of E.coli,Staphylococcus aureus and Salmonella were 6.25 mg / m L,12.5mg / m L,25 mg / m L,respectively;and the lowest values for MBC of E.coli,Staphylococcus aureus and Salmonella were 12.5 mg / m L,25 mg / m L and 50 mg / m L,respectively.The results showed that compound pulsatilla powder had good antimicrobial affect against E.coli,Staphylococcus aureus and Salmonella.1.2 Study on analgesic anti-inflammatory activity.The analgesic and anti-inflammatory effects of compound pulsatilla powder were measured by the methods of acetic acid-induced writhing,hot plate and xylene induced auricle swelling in mice.The analgesic results showed that both moderate and high dose modified powder significantly increased the pain threshold and reduced the number of writhing reactions induced by acetic acid in mice.The inhibitory rate of high dose on writhing reaction in mice reached to 61.77%,with a significant difference compared with the model group(P < 0.05).The anti-inflammatory results showed that the inhibition rate of compound pulsatilla powder on auricle swelling of mice induced by xylene was 80.77%,with a significant difference compared with the model group(P < 0.05).The results showed that compound pulsatilla powder had anti-inflammatory and analgesic effect.2 Study on the antidiarrheal effect of compound pulsatilla powderThe antidiarrheal effect of the compound pulsatilla powder was studied in mice model induced by castor oil,and the effects of compound pulsatilla powder on gastric emptying and intestinal propulsion in mice were studied by semisolid nutritional paste method.The isolated rabbit smooth muscle ileum was to observe the effect of compound pulsatilla powder on spontaneous contraction of rabbit smooth muscle ileum in normal condition.The results showed that in the model of acute diarrhea induced by castor oil,compound pulsatilla powder could significantly inhibit the defecation times of wet faeces in mice within 6 hours after conducting with castor oil(P < 0.05).High dose of compound pulsatilla powder significantly decreased the intestinal propulsive rate and increased gastric emptying rate of normal mice(P < 0.05).Compound pulsatilla powder inhibited spontaneous contraction of isolated rabbit small intestinal smooth muscle ileum in a concentration-dependent manner(P < 0.05).The results showed that compound pulsatilla powder had a good antidiarrheal effect and could significantly inhibit the small intestine movement under normal conditions.3 The effect of compound pulsatilla powder on diarrhea induced by E.coli in miceThe mice were selected to establish E.coli diarrhea model,continuous administration by gavage for 5 days,to observe and record the weight and the diarrhea index.Through the detection of number of leukocyte;weight determination of immune organs;detection of IL-6 and TNF-? cytokine levels by Elisa method;the determination of intestinal microflora number by the plate counting method,evaluate the effect of compound pulsatilla powder on the mice with diarrhea induced by E.coli.The results show that,(1)Compared with the model group,the moderate and high dose of compound pulsatilla powder significantly increased the body weight and decreased the diarrhea index of diarrhea mice(P < 0.05).(2)Compared with the model group,the moderate and high dose of compound pulsatilla powder significantly decreased the leukocyte count of diarrhea mice,and the levels of cytokine IL-6 and TNF-alpha were significantly decreased(P < 0.05).(3)Compared with the model group,the spleen and thymus index of the moderate and high dose group increased significantly(P < 0.05),the number of probiotics in caecum contents of mice was significantly increased and the number of harmful bacteria decreased(P < 0.05).These results suggest that compound pulsatilla powder can improve the body weight,decrease the diarrhea index,reduce the inflammatory reaction,restore immunity and regulate the balance of intestinal microflora in diarrhea mice.Compound pulsatilla powder has a good therapeutic effect on diarrhea and can improve the complications of diarrhea.
