| Rotavirus can cause diarrhea and death in infected piglets.It is known that Lactobacillus amylovorus,Radix pulcherum powder and zinc oxide all have corresponding prevention and treatment effects on diarrhea infection.In order to improve the prevention and treatment effect,MA104 cells were selected to evaluate the effect of Lactobacillus amylovorus.The neonatal piglets were purchased from milk mice in Kunming,China and a pig farm in Harbin to evaluate the efficacy of three preparations combined to improve the prevention and treatment of rotavirus infection.Mainly adopts CCK-8 method,ELISA method and PCR method,HE dyeing method,by looking at different incubation time mixed lactobacillus food starch suspension to prevent rats RV infection,food starch lactobacillus,silver and zinc oxide in the prevention and treatment of clinical symptoms of rats infected with RV,growth performance,spleen index,the jejunum intestinal villus,detoxification and detoxifying the change of time,And explore the damage of RV titer in duodenum and jejunal villi.To compare the prevention and treatment effects of Lactobacillus amylovorus,Pulsatilla chinensis powder and zinc oxide on diarrhea caused by rotavirus infection in Suckling mice and piglets.The experimental results showed that the supernatant of Lactobacillus amylovorus cultured for 16 h and 21 h significantly inhibited RV,and the difference was not significant.The effect of Lactobacillus amylovorus combined with P.pulsatilla(50mg/ m L)and zinc oxide(2.5mg/ m L)on preventing RV infection in suckled mice was better than that of Lactobacillus amylovorus combined with P.pulsatilla(100mg/ m L),zinc oxide(10mg/ m L)and zinc oxide(5mg/ m L).The weight change and growth rate of Lactobacillus amylopeculis combined with Baikoutou powder and zinc oxide to prevent RV in Suckling mice were lower than those in normal group.After 5-10 days,except for the normal group,the weight gain rate of probiotics + P.pulsatilla was the highest,followed by probiotics + zinc oxide.The lowest diarrhea rate was 0.00% in the group infected with5 d probiotics + P.pulsatillum and 25% in the group infected with virus.There was no significant difference in spleen index between probiotics and probiotics on day 3 of infection(P<0.05),but spleen index on day 5 and 7 was significantly higher than that in other groups(P<0.05).Morphological changes of jejunum showed that there was no significant difference in villus damage between probiotics and probiotics and zinc oxide in the jejunum of neonatal mice,and the effect was better than that of single use.ELISA results showed that the RV content in the combination group was significantly lower than that in the single group.PCR results showed that 10 days after infection with RV,the virus scavenging rate of the virus group was 60%,the virus scavenging rate of the probiotics was 80%,and the virus scavenging rate of the other groups was100%.The results of small intestine cell activity indicated that the cell activity of the combination group was better than that of the single group.The combination of Lactobacillus amylogolum,Pulsatilla pulsatilla powder and zinc oxide was used to treat RV of Suckling mice.On the 9th day of feeding,the body weight of Suckling mice was the highest in the normal group,followed by probiotics + Pulsatilla pulsatilla,and the lowest in the virus group.The diarrhea ratio of probiotics+ pulsatillum pulsatillum infected for 5 days was 0.00%,and the diarrhea ratio of probiotics + zinc oxide was 9.09%.The diarrhea ratio was better than that of the single drug group.On day 7 of RV infection,spleen index of probiotics + zinc oxide was higher than that of virus group and normal group(P<0.05),but there was no significant difference among other groups(P>0.05).Morphological changes of jejunum showed that there was no significant difference in villus damage between probiotics and probiotics and zinc oxide in the jejunum of neonatal mice,and the effect was better than that of single use.ELISA results showed that the content of RV in suckled mice decreased gradually after infection,and the content of RV in probiotics + P.pulsatilis group for 7 days was the lowest.In the treatment of piglets RV with Lactobacillus amylopeculis combined with P.chinensis powder and zinc oxide,the morphological changes of jejunum showed no significant difference in villus damage between probiotics + P.chinensis and probiotics + zinc oxide,and the effect was better than that of single use.RV titer test showed that probiotics +pulsatilla chinensis and zinc oxide could reduce RV replication.In summary,the results showed that the suspension of Lactobacillus amylovorusw ith different culture time had no significant difference in preventing RV infection in Suckling mice.Lactobacillus amylopeculis mixed with Baitouweng powder(50mg)and zinc oxide(2.5mg)could improve the growth performance,reduce the proportion of diarrhea,improve spleen index,increase the activity of intestinal cells in small intestine,reduce the damage of intestinal villus in jejunum,and reduce the amount of detoxification and the time of detoxification.At the same time,Lactobacillus amylovorus combined with Baikongwong powder and zinc oxide prophylactic use can reduce RV replication,and reduce jejunal villus injury. |
