| The experiment was in the diet that supplemented separately with 0.8%CA-OKGM,0.8%CA-OKGM Complex1 and 0.8%CA-OKGM Complex2 to feed Schizothorax prenanti for 8 weeks,and then measured the growth index of fish,the immune index and antioxidant index in serum,the expression of immune-associated genes in the head kidney,spleen,liver and midgut;It also measured the immune index and antioxidant index in serum,the expression of immune-associated genes in the head kidney,spleen,liver and midgut after the fish infected with Aeromonas hydrophila in 6h,72 h,24h,and 168 h,and on this account to explore the effect of CA-OKGM and its complexs on the immune performance and related gene expression of the fish in Schizothorax prenanti.This article mainly involved the following three aspects:1?The effect of CA-OKGM and its complexs on the growth performance in Schizothorax prenanti.CA-OKGM and its complexes could significantly improved the weight gain and specific growth rate of Schizothorax prenanti,and the effect of CA-OKGM Complex1 and CA-OKGM Complex2 was better than that of CA-OKGM,but all had no significant effect on the immune organ index.2?The effect of CA-OKGM and its complexs on the immune index and antioxidant index in the serum of Schizothorax prenanti.Immunization:(1)in normal aquaculture experiments,CA-OKGM Complex1 and CA-OKGM could significantly increase ACH50 activity and CRP,C3 and Ig M contents in the serum of the fish,and the Ig M content increased by 2.19 times and 2.73 times respectively.CA-OKGM Complex2 could only significantly increase the content of Ig M,which is 1.77 times as high as that of the base group,and there was no significant difference in other indicators.(2)In the attack test,CA-OKGM Complex1 and CA-OKGM Complex2 could both rapidly promote the generation of humoral immune factors in serum,and regulate the immune response,but CA-OKGM Complex1 had better immunoregulation effect.Antioxidation:(1)in normal aquaculture experiments,CA-OKGM and its complexes showed better antioxidant function,CA-OKGM and CA-OKGM Complex1 significantly increased the activity of GSH-PX and T-SOD in the serum of the fish,and the two activities in CA-OKGM Complex1 respectively were the highest up to 207.49±1.05U/m L and 477.45±8.72 U;The two material could significantly reduced the content of MDA in serum,too.CA-OKGM Complex2 also significantly increased T-SOD activity,significantly decreased MDA content,and increased GSH-PX activity with no significant effect.CA-OKGM Complex1 could significantly reduce the activity of i NOS and the content of NO,but CA-OKGM and CA-OKGM Complex2 could significantly increase the activity of i NOS and reduce the content of NO.(2)In the attack test,CA-OKGM and CA-OKGM Complex1 showed better antioxidant function,which could improve the antioxidant enzyme activity,reduce free radicals,inhibit peroxide production and resist oxidative damage to varying degrees.3?The effect of CA-OKGM and its complexs on the expression of immune-associated genes in the tissues of Schizothorax prenanti.(1)In normal aquaculture experiments,CA-OKGM Complex1 could improve the expression of tlr5 to different degrees in different tissues of Schizothorax prenanti and increase the expression of tlr22 in spleen and midgut;CA-OKGM and CA-OKGM Complex2 could improve the expression of tlr5 and tlr22 in spleen and midgut;Each test group could improve the expression of ?2-integrin in immune organ tissues.Both CA-OKGM Complex2 and CA-OKGM Complex1 could increase the expression of cd40 and pkr in the head kidney,spleen and liver;while CA-OKGM reduced the expression of cd40 and pkr in the head kidney but increased the expression in spleen and liver.CA-OKGM Complex2 can enhance the expression of myd88 in head kidney,spleen and liver;And CA-OKGM Complex1 in the same immune organs like CA-OKGM Complex2 decreased the expression of My D88;CA-OKGM significantly decreased the expression of My D88 in kidney and spleen and significantly increased the expression in the liver.CA-OKGM Complex1 could significantly improve the relative expression of traf6,irf7 and nf-kbp100 in head kidney,liver,spleen and midgut;CA-OKGM Comple2 and CA-OKGM could only increase the expression of traf6,irf7 and nf-kbp100 in part of the immune tissues,while the expression of TRAF6 and IRF7 in the liver were significantly decreased.All the experimental groups could reduce the expression of il-1 and il-8 in the immune tissue to varying degrees.For tnf-?,CA-OKGM could significantly increase its expression in the spleen and midgut,and CA-OKGM Complex1 could significantly increase their expression in the liver and midgut.Each test group could increase the expression of il-10 and tgf-? in immune tissues;for hsp70,CA-OKGM Complex1 could significantly improve its expression in spleen,liver and head kidney,and CA-OKGM Complex2 and CA-OKGM could significantly improve the expression in liver in different degree and decrease its expression in head kidney,spleen and liver.(2)In the attack test,by Aeromonas hydrophila infecting the fish,TLR5 signaling pathway was activated first,while the increase in expression of tlr22 was in late stage of bacterial infection;all genes' expressions would not peak at the same time points.however,they would be more concentrated in a certain period of time.The expression of immune factors in the head and midgut is very fast,especially in the head kidney.CA-OKGM and CA-OKGM Complex1 can regulate the expression of immune factors in the head kidney and midgut,regulate the immune response and promote the expression of tlr22 in the head kidney and intestines.In liver and spleen,CA-OKGM Complex2 and CA-OKGM can better regulate their cytokine's expression.CA-OKGM Complex1 shows good immunoregulatory ability in the early stage. |
