Cervical cancer is not only the fourth most common malignant tumor in the world,but also one of the leading causes of cancer death in women.The 3-to 5-year survival rate of cervical cancer in many underdeveloped countries is < 50%.Although chemotherapy or neoadjuvant chemotherapy has been used for the treatment of cervical cancer,the toxicity and resistance of chemotherapy to the human body is obvious.It has long been established that antioxidants and other active compounds from edible and medicinal fungi may help prevent cancer,and natural compounds from edible and medicinal fungi may be considered as complementary drugs in the treatment of cervical cancer.In recent years,some new varieties of mushroom genus have been continuously discovered and reported.Agaricus sinodeliciosus is a new species reported in recent years in the genus Mushroom.It is a huge mushroom and has been domesticated and cultivated successfully.Finding anti-tumor active substances can solve the problem of insufficient raw materials for drugs to a large extent.The experimental material of this study is a huge wild mushroom collected from Xinjiang-" Agaricus sinodeliciosus ".In this paper,we first study the biological characteristics,domestication cultivation and nutrient composition of Agaricus sinodeliciosus mycelium by using Agaricus sinodeliciosus mycelium as research material.Secondly,the petroleum ether extraction was evaluated by the crude extracts of Agaricus sinodeliciosus wild fruit bodies through cell migration,cloning,cell proliferation,apoptosis,release of ROS,expression of PARP and expression of apoptosis-related proteins.And anti-cervical cancer Hela cell activity of chloroform extract,and further explore the mechanism of action of Agaricus sinodeliciosus extract,The study found that the extract of Agaricus sinodeliciosus wild fruit body has a good inhibitory effect on Hela cells.However,the Agaricus sinodeliciosus extract is a mixture with many active substances of unknown composition,but it is difficult to find the monomeric substances that play a major active role.Therefore,the petroleum ether extract was isolated and identified,and a monomeric compound ergosterol was isolated and evaluated for anti-cervical cancer activity.The main results are as follows:1.Glucose is the most suitable carbon source for the growth of Agaricus sinodeliciosus hyphae.Soy peptone is the most suitable nitrogen source for the growth of Agaricus sinodeliciosus hyphae;the most suitable temperature for the growth of Agaricus sinodeliciosus hyphae is 25 °C,Chinese mushroom mushroom The optimum p H for silk growth is 6.The preliminary domestication showed that the average yield of the first mushroom of the reed cultivation substrate was 0.245 kg per rod,and the biological efficiency was 38.5%.Agaricus sinodeliciosus are not only high in protein,low in fat,rich in dietary fiber and minerals,but also have high health value.It is a rare dual-use drug.2.Both petroleum ether extract and chloroform extract can inhibit the migration of Hela cells,clone formation,cell proliferation,promote cell apoptosis,and promote PARP protein expression and ROS release.Its mechanism of inhibiting the proliferation of cervical cancer Hela cells is mainly achieved by up-regulating the expression of Bax,Bad,P53,AIF and cleaved-caspase-3 proteins and down-regulating the expression of Bcl-2 and pro-capase-3 proteins.3.HPLC and NMR methods were used to identify the anti-tumor active ingredients of Agaricus sinodeliciosus petroleum ether extract and chloroform extract.Component A in the petroleum ether extract was identified as ergosterol.Cell proliferation experiments showed that petroleum ether extract and ergosterol had strong inhibitory effects on Hela cells,and petroleum ether extract inhibited Hela cells more than ergosterol.This study mastered the biological characteristics of Agaricus sinodeliciosus and successfully cultivated them.At the same time,the anti-tumor activity of Chinese wild mushroom wild fruit extracts was found to be better for Hela and petroleum ether extracts and chloroform extracts.Inhibition,its mechanism of action is mainly regulated by promoting apoptosis.An active substance was isolated from petroleum ether extract and identified as ergosterol,and ergosterol also inhibited the increase of Hela cells.It can be seen that this study can provide a scientific basis for the future commercial cultivation of Agaricus sinodeliciosus,and provide a theoretical basis for the research on the anti-tumor activity of Agaricus sinodeliciosus extracts.The active ingredients of Agaricus sinodeliciosus have the potential to develop into potential chemotherapy drugs for the treatment of cervical cancer.
