In this experiment,eight kinds of Chinese herbal medicine(Codonopsis pilosula,Largehead atractylodes rhizome,Poria cocos and Liquorice etc.)were processed into supplementary SIJUNZI decoction and added into the diet of canines in order to study the effects of supplementary SJZD on the expression levels of Epidermal growth factor(EGF)and Somatostatin(SS),and their mRNA relative expression values in small intestine of canines,intestinal digestive and absorptive function,immune function in canines,and further to explore mechanism of different dosages of supplementary SJZD on effect of digestive and Immune function in canines,to provide experimental basis for its clinical application.Eighteen one-year-old beagles were selected and randomly divided into three groups,six canines per group.The three groups were group A(control group),group B(1%supplementary SJZD group),group C(2%supplementary SJZD group).Canines in group A were fed basal diets.In group B and group C,1%and 2%supplementary SJZD were added to the basal diet,respectively.Experiment period was fourteen days.The blood samples were taken at the 0,7,14 d,and the serum levels of D-xylose,Interfern gamma(IFN-?),Interleukin-2(IL-2),Interleukin-4(IL-4),Interleukin-6(IL-6),Immunoglobulin A(IgA),Immunoglobulin G(IgG),Immunoglobulin M(IgM)were detected by enzyme linked immunosorbent assay(ELISA).On the 14 d,all the canines were killed for collecting pancreas,duodenum,jejunum and ileum,the digestive enzyme activities in pancreas were measured by ELISA;the protein positive expression levels of EGF,SS,Secretory immunoglobulin A(SigA)in small intestine were measured by Immunohistochemistry(IHC),and the relative expression value of EGF,SS,IgA,IgG,IgM mRNA in small intestine were measured by Real-time Polymerase Chain Reaction(RT-PCR).The results were as follows:1.At 7,14 d,compared with group A and group B,the serum D-xylose contents of canines in group C were significantly increased(P<0.05);compared with group A,the serum D-xylose contents of canines in group B were not significantly difference(P>0.05).2.Compared with group A and group B,the digestive enzyme activities in pancreas of canines in group C were significantly increased(P<0.05);compared with group A,the digestive enzyme activities in pancreas of canines in group B were not significantly difference(P>0.05).3.Compared with group A,at 7 d,the serum IFN-,y,IL-2,IgA,IgM levels of canines in group C were significantly increased(P<0.05),levels of IgG were significantly increased(P<0.01);at 14 d,the serum IFN-? levels of canines in group B were significantly increased(P<0.05),the serum IFN-y,IL-2,IL-6,IgM levels of canines in group C were significantly increased(P<0.05),levels of IgA and IgM were significantly increased(P<0.01).4.Compared with group A,the protein positive levels of EGF in small intestine of canines in group B and group C were significantly increased(P<0.01).Compared with group B,the protein positive levels of EGF in duodenum and jejunum of canines in group C were significantly increased(P<0.01),in ileum were significantly increased(P<0.05).Compared with group A,the EGF mRNA relative expression values in jejunum of canines in group B were significantly increased(P<0.05),in ileum were significantly increased(P<0.01),the EGF mRNA relative expression values in small intestine of canines in group C were significantly increased(P<0.01)Compared with group B,the EGF mRNA relative expression values in duodenum and jejunum of canines in group C were significantly increased(P<0.01).5.Compared with group A,the protein positive levels of SS in duodenum of canines in group B were significantly decreased(P<0.01),in ileum were significantly decreased(P<0.05),the protein positive levels of SS in small intestine of canines in group C were significantly decreased(P<0.01).Compared with group B,the protein positive levels of SS in small intestine of canines in group C were significantly decreased(P<0.01).Compared with group A,the SS mRNA relative expression values in duodenum and ileum of canines in group B were significantly decreased(P<0.01),in jejunum were significantly decreased(P<0.05),the SS mRNA relative expression values in small intestine of canines in group C were significantly decreased(P<0.01).Compared with group B,the SS mRNA relative expression values in duodenum of canines in group C were significantly decreased(P<0.01),in jejunum and ileum were significantly decreased(P<0.05).6.Compared with group A,the protein positive levels of SIgA in duodenum of canines in group B were significantly increased(P<0.01),jejunum and ileum were significantly increased(P<0.05),the protein positive levels of SIgA in duodenum andjejunum of canines in group C were significantly increased(P<0.01),in ileum were significantly increased(P<0.05).Compared with group B,the protein positive levels of SIgA in jejunum of canines in group C were significantly increased(P<0.05).Compared with group A,the SIgA mRNA relative expression values in small intestine of canines in group B and group C were significantly increased(P<0.01).Compared with group B,the protein positive levels of SIgA in duodenum and jejunum of canines in group C were significantly increased(P<0.01).7.Compared with group A,the IgG mRNA relative expression values in duodenum and jejunum of canines in group B were significantly increased(P<0.05),the IgG mRNA relative expression values in small intestine of canines in group C were significantly increased(P<0.01).Compared with group B,the IgG mRNA relative expression values in jejunum and ileum of canines in group C were significantly increased(P<0.05).Compared with group A,the IgM mRNA relative expression values in duodenum and jejunum of canines in group B was significantly increased(P<0.05),the IgM mRNA relative expression values in duodenum and jejunum of canines in group C were significantly increased(P<0.01),in ileum were significantly increased(P<0.05).Compared with group B,the IgM mRNA relative expression values in duodenum of canines in group C were significantly increased(P<0.05).In conclusion,this experimental results suggested that supplementary SJZD could improve the intestinal absorptive and immunology function,increase the digestiveenzyme activities of pancreas and levels of humoral immunity,enhance digestive and immune function of canines.The effects of 2%supplementary SJZD were prior to 1%supplementary SJZD.
