| Mucuna macrocarpa Wall.is a large woody vine plant distributed in Guangdong,Guangxi,Yunnan,Guizhou and other places.Mucuna macrocarpa Wall has good prospects for garden application.Its cane can be used for medicinal purposes.It is often used to strengthen the body,regulate menstruation,supplement blood,treat sequelae of poliomyelitis,and treat rheumatoid bone pain.Methyl Jasmonate(MeJA)and salicylic acid(SA)have effects on plant physiological activities and plant metabolic activities.In this study,Mucuna macrocarpa Wall,was sprayed with 0.1 mmol/L,0.5 mmol/L,0.9 mmol/L,1.3 mmol/L MeJA and SA.By comparing the growth,physiology,chlorophyll fluorescence parameters,medicinal components and other indicators of Mucona macrocarpa Wall,to determine the types and concentrations of preferred growth regulators suitable for Mucuna macrocarpa Wall growth,explore the effects of growth regulators on the nutrient content and secondary metabolite accumulation of medicinal vines,and provide a theoretical basis for improving the cultivating level of vines.The main findings are as follows:(1)MeJA has a slight inhibitory effect on the stem of Mucona macrocarpa Wall,while low concentration of SA promotes the growth of stems,and the concentration is too high to produce inhibition.Treatment with 0.9 mmol/L and 1.3 mmol/L methyl jasmonate promoted the increase of leaf area of Mucuna macrocarpa Wall.Under the treatment of 0.1mmol/L salicylic acid,the soil diameter increased,the biomass and leaf area were the largest,which was most beneficial to promote plant growth and biomass accumulation.The leaf area and biomass of Mucuna macrocarpa Wall were strongly correlated with the content of indole acetic acid IAA,zeatin nucleoside ZR and gibberellin GA3.(2)Low concentration of MeJA and SA treatment can increase the activity of SOD,POD and CAT,while over a certain concentration,it shows the inhibition of its activity.It can be seen that exogenous application of 0.5 mmol/L methyl jasmonate and salicylic acid It can greatly increase the activity of SOD,POD and CAT,thus helping to eliminate the toxic effects of H2O2 in cells.The highest content of soluble sugar and soluble protein in MJ-1,MJ-4 treatment is beneficial to improve cell osmotic adjustment ability.(3)Different concentrations of methyl jasmonate and salicylic acid are bene:ficial to increase the content of chlorophyll a and chlorophyll b in Mucuna macrocarpa Wall.The effective photochemical rate(Fv’/Fm’),non-photochemical quenching coefficient(NPQ),electron transport rate(ETR)and photosynthetic quenching coefficient(qP)under salicylic acid treatment were slightly lower than methyl jasmonate treatment.Treatment with 1.3mmol/L methyl jasmonate is more conducive to increase the maximum photochemical rate(Fv/Fm),Fv’/Fm’,qP,ETR,and lower NPQ,which is more conducive to improving effective light energy and photosynthetic activity.(4)Indole acetic acid IAA content,zeatin nucleoside ZR content,gibberellin GA3 content,the ratio IAA/ABA and(IAA+GA3+ZR)/ABA of Mucona macrocarpa Wall treated with 1.3 mmol/L methyl jasmonate and 0.1 mmol/L and 0.5 mmol/L salicylic acid is higher,which is beneficial to the increase of endogenous hormone levels in Mucuna macrocarpa Wall.MeJA and SA reduce the abscisic acid ABA content in plant leaves.(5)The various nutrient contents are closely related to total phenol,total flavonoids and total triterpenic acid.In general,the treatment of methyl jasmonate at each concentration was beneficial to increase the accumulation of Fe and Mn in Mucuna macrocarpa Wall.0.1mmol/L salicylic acid can promote the absorption of Ca element by Mucana macrocarpa Wall.0.1 mmol/L and 1.3 mmol/L methyl jasmonate are beneficial to the absorption of Mg by various organs.0.1mmol/L,1.3mmol/L salicylic acid and 0.5mmol/L methyl jasmonate treatment are most beneficial to the accumulation of total phenol in plant roots The accumulation of total phenol in stems of Mucona macrocarpa Wall was most significant with 1.3 mmol/L salicylic acid.Medium concentration(0.5mmol/L,0.9mmol/L)of MeJA and 0.5mmol/L salicylic acid treatment promoted the accumulation of total phenol in leaves.The accumulation of total flavonoids in stems and leaves under MeJA treatment was slightly higher than that in SA treatment.Comprehensive analysis,low concentration of MeJA and high concentration of SA treatment is beneficial to the accumulation of total flavonoids and total triterpenoids in Mucuna macrocarpa Wall(6)The effects of two growth regulators on the coefficient(qP),IAA/ABA,(IAA+GA3+ZR)/ABA,and significantly positively correlated with ZR content.Leaf area was extremely significantly positively correlated with qP,IAA/ABA,(IAA+GA3+ZR)/ABA,and was significantly positively correlated with maximum photochemical rate Fv/Fm,IAA content and ZR content. |
