| Mongolian sheep production is increasing rapidly.As a result,sheep fat production also increased.At present,sheep fat in our country can not be fully utilized.It leads to the accumulation and waste of sheep fat.Therefore,the comprehensive utilization of sheep fat needs to be further studied.This experiment uses 6,18,30 months Sunite sheep as the research object,take the tail fat,perirenal fat,skeletal muscle fat after slaughter.High-throughput data was obtained using UHPLC-Q-TOF/MS from Sunite sheep adipose tissue samples.Identification of metabolites by metabolite primary m/z identification and secondary mass spectrometry.The multivariate statistical method was used to observe the grouping trend of different samples.Inter-group differential metabolites were screened in these conditions:FC≥2 or FC≤0.5;p-value≤0.05;VIP≥1.0.To explore the differences of lipid metabolites of tail fat,perirenal fat and skeletal muscle fat in sunit sheep at different months,and differences between lipid metabolites of tail fat and perirenal fat and skeletal muscle fat at different growth stages,To provide theoretical basis for comprehensive utilization of sheep fat.The result showed as follows:Detection of adipose tissue samples from Sunite sheep,4283 ion information was extracted in positive ion mode,and 2722 ion information was extracted in negative ion mode.Obtained 171 high-confidence lipid metabolites,Special polyunsaturated long carbon chains C20:4,C20:5,C22:4,C22:5 were also found in the phospholipids of Sunite sheep’sadipose tissue.There were significant differences in the levels of glycerolipids,sphingomyelins,glycerophospholipids and fatty acyls at 6 months,18 months,and 30 months of age in sunit sheep fat,perirenal fat and skeletal muscle fat(p<0.05).Decreased triglyceride in tail fat and skeletal muscle fat with age(p<0.05).Glypholipids and sphingomyelins in 18 months tail fat and skeletal muscle fat were higher than those at 6 months of age(p<0.05).The oleic acid and diglyceride in the 30 months tail fat were higher than 6 months and 18 months(p<0.05).glycerophospholipids and sphingomyelins in the perirenal fat decreased with age(p<0.05).There were significant differences in glycerolipids,glycerophospholipids,fatty acyls and sphingomyelin between tail fat,perirenal fat and skeletal muscle fat of 6,18 and 30 months Sunit sheep(p<0.05).The contents of diacylglycerol(DG)and oleic acid in tail fat were significantly higher than those in skeletal muscle fat and perirenal fat(p<0.05),and the contents of glycerophospholipid and sphingomyelin in tail fat at different months were higher than those in skeletal muscle fat and perirenal fat. |
