In this experiment,the aim is to understand the expression rules of Stearpyl-CoA desaturase(SCD gene)in blood of Sunite sheep of different populations and the correlation between SCD gene and the composition and content of fatty acids.On the basic of a genetic perspective,to verify the root cause of Sunite sheep's unique flavor and high nutritional value.The test collected 20 parts of the 3-year-old Sunite male and female sheep blood from the grazing as test materials.Gas chromatography was used to determine the composition and content of fatty acids in samples.SCD gene was regarded as the target gene.Fluorescence quantitative PCR technology was used to detect and calculate the SCD gene expression in samples.The correlation between SCD gene expression and fatty acid composition was analyzed.Finally,the differences in DNA sequenes between male and female were analyzed.The results of the experiment showed that there was no difference in composition of fatty acid between Sunite male and female sheep.However,the detected content of fatty acid had a number of difference.Compared with female sheep,the content of saturated fatty acids in male sheep was lower.However,the contents of monounsaturated fatty acid and polyunsaturated fatty acid were relatively high.The content of stearic acid causing smell of mutton was low;The oleic acid content in monounsaturated fatty acids was high regardless of male and female sheep.Moreover,the oleic acid content in monounsaturated fatty acid of male sheep was higher than that of female sheep.As a result,it could be inferred that male sheep had better flavor and higher nutritional value than female sheep.There was bigger difference in the expression of SCD gene between male and female sheep.The results of 6 samples in the male sheep were higher than that in the female sheep.Only results of 4 samples in the female sheep were higher than that in the male sheep.The expression of male sheep was significantly higher than female sheep.The results of SCD gene expression and fatty acid correlation in male and female sheep showed that the expression of SCD gene mRNA in blackhead male was significantly positively correlated with oleic acid,and the expression of SCD gene mRNA in blackhead female was not correlated with the detected fatty acids.On the basic of comparison and analysis of the DNA sequences of male and female sheep,it was found that the similarity of DNA sequences has reached ninety-three point eight five percent.
